Host and gut microbial tryptophan metabolism and type 2 diabetes: an integrative analysis of host genetics, diet, gut microbiome and circulating metabolites in cohort studies

Qibin Qi et al.

Summary: The study found positive associations between tryptophan and its metabolites with the risk of type 2 diabetes, while indolepropionate was inversely associated with the risk. Additionally, host genetic variants, dietary factors, gut bacteria, and their interplay related to these type 2 diabetes-related metabolites were identified.

Dietary cholesterol drives fatty liver-associated liver cancer by modulating gut microbiota and metabolites

Xiang Zhang et al.

Summary: High dietary cholesterol leads to sequential progression of steatosis, steatohepatitis, fibrosis and eventually HCC in mice, concomitant with insulin resistance. Cholesterol-induced NAFLD-HCC formation is associated with gut microbiota dysbiosis, with specific bacterial composition changes between stages of liver disease. Alterations in gut bacterial metabolites were also observed with dietary cholesterol intake. Therapies targeting cholesterol and gut microbiota may be effective for preventing NAFLD-HCC.

The Aryl Hydrocarbon Receptor in Energy Balance: The Road from Dioxin-Induced Wasting Syndrome to Combating Obesity with Ahr Ligands

Nathaniel G. Girer et al.

Summary: The aryl hydrocarbon receptor (AHR) is a ligand-activated transcription factor that plays a crucial role in various physiological processes, including organ development, immune system function, wound healing, and energy metabolism. Recent studies have suggested that AHR could be a potential therapeutic target for obesity.

Indole-3-propionic Acid Improved the Intestinal Barrier by Enhancing Epithelial Barrier and Mucus Barrier

Jiaojiao Li et al.

Summary: The study demonstrated that indole-3-propionic acid (IPA) can enhance intestinal barrier function by increasing the expression of tight junction and mucin proteins, while decreasing inflammatory factors. This suggests a potential role for gut microbial metabolites of aromatic amino acids in improving intestinal barrier integrity.

High-fat diet-induced upregulation of exosomal phosphatidylcholine contributes to insulin resistance

Anil Kumar et al.

Summary: This study reveals that lean mice become insulin resistant after being administered exosomes isolated from the feces of obese mice fed a high-fat diet or from patients with type II diabetes. The altered lipid composition of exosomes from high-fat diet-fed mice affects the uptake by macrophages and hepatocytes, leading to inhibition of the insulin signaling pathway. Exosome-derived phosphatidylcholine activates AhR, inhibiting the expression of genes essential for activation of the insulin signaling pathway.
