Efficient one-pot, three-component procedure to prepare new α-aminophosphonate and phosphonic acid acyclic nucleosides

Summary: The one-pot three-component Kabachnik-Fields reaction using aldehydes, amines, and triethyl phosphite proceeded efficiently with natural phosphate coated with iodine as a catalyst. The synthesized alpha-aminophosphonates and phosphonic acid acyclic nucleosides showed poor anti-HCV and anti-HIV activities possibly due to the absence of hydrophobic pharmacophores.

Design, Synthesis, and Activity Evaluation of Novel Acyclic Nucleosides as Potential Anticancer Agents In Vitro and In Vivo

Summary: The study designed, synthesized, and evaluated 103 novel acyclic nucleosides for their anticancer activities, with compound 9b showing promising inhibitory effects on colon cancer cells. All (R)-configured derivatives displayed stronger anticancer activity, with compound 9b inducing apoptosis and inhibiting colony formation. Importantly, compound 9b exhibited low systemic toxicity and inhibited tumor growth in a mouse model, suggesting it as a potential lead compound for future anticancer drug discovery.

Palladium-catalyzed asymmetric allylic amination of a vinylethylene carbonate with N-heteroaromatics†

Summary: A highly enantioselective allylation reaction of vinylethylene carbonate by asymmetric palladium catalysis has been developed for various N-heteroaromatic substrates. The efficient synthesis of acyclic nucleoside analogs with high yields (up to 96%) and enantioselectivities (up to 99%) was demonstrated with only 0.5 mol% of Pd catalyst. Additionally, these acyclic nucleosides can undergo rapid transformation into a variety of structurally diverse chiral acyclic nucleosides and isonucleosides.
