Hippocampal neural stem cells and microglia response to experimental inflammatory bowel disease (IBD)

Ioannis-Alexandros Gampierakis et al.

Summary: This study investigates the effects of acute and chronic experimental colitis on adult hippocampal neurogenesis and innate immune cell responses. It reveals that acute colitis enhances neurogenesis but with deficits in cell cycle kinetics, while chronic colitis shows normal levels of neurogenesis but deficits in neuron migration and integration. Additionally, acute colitis leads to increased inflammation in the hippocampus, while chronic colitis results in elevated levels of tissue-repairing cells and anti-inflammatory cytokines. These findings highlight potential mechanisms underlying cognitive and mood dysfunction in patients with IBD.

Inflammatory bowel disease and Parkinson's disease: common pathophysiological links

Ho-Su Lee et al.

Summary: Inflammatory bowel disease and Parkinson's disease have been shown to have a bidirectional link, with shared genetic loci such as LRRK2. Recent studies suggest that inflammatory bowel disease may increase the risk of developing Parkinson's disease. The underlying mechanisms of gut-brain interactions are still not fully understood, but recent findings are beginning to shed light on this connection.

The interplay between Glucocerebrosidase, α-synuclein and lipids in human models of Parkinson's disease

Sonia Sanz Munoz et al.

Summary: Mutations in the GBA gene, encoding GCase, are the highest genetic risk factor for Parkinson’s disease, leading to decreased GCase activity and stability which results in the accumulation of lipid substrates and αS. Changes in αS and lipid levels are correlated with alterations in GCase activity/protein levels. Current strategies aim to revert the changes in GCase activity/protein, αS, and lipid levels in the context of PD.

The impact of dextran sodium sulphate and probiotic pre-treatment in a murine model of Parkinson's disease

Zach Dwyer et al.

Summary: This study examined the effects of pre-treatment with a probiotic and an inflammatory inducer on PD pathology in a murine model. The probiotic showed no significant effects, while the inflammatory inducer caused marked changes in gut microbiome and increased behavioral and inflammatory outcomes. These findings suggest a potential link between disruption of intestinal integrity and systemic inflammatory events in promoting brain-gut changes relevant to PD.

Brain Microglial Activation Increased in Glucocerebrosidase (GBA) Mutation Carriers without Parkinson's disease

Stephen Mullin et al.

Summary: The study found that carriers of glucocerebrosidase gene mutations without Parkinson's disease also present neuroinflammation. Microglial activation was observed in brain regions susceptible to Lewy pathology. Importantly, the degree of hyposmia was positively correlated with nigral binding potential.

Summary: A retrospective study indicates that patients with irritable bowel syndrome (IBS) have an increased risk of developing Parkinson's disease (PD) in the first two years, but not in the long term. The findings suggest that IBS may be a prodromal symptom of PD rather than an actual risk factor.

Glucocerebrosidase reduces the spread of protein aggregation in a Drosophila melanogaster model of neurodegeneration by regulating proteins trafficked by extracellular vesicles

Kathryn A. Jewett et al.

Summary: This study shows that GCase deficiency leads to abnormal protein aggregation within EVs, and restoring GCase protein reduces protein aggregation and affects the spread of protein aggregation between tissues. These findings are important for understanding PD progression and may lead to new targets for slowing neurodegeneration.

Schizophrenia-associated SLC39A8 polymorphism is a loss-of-function allele altering glutamate receptor and innate immune signaling

Wei Chou Tseng et al.

Summary: The A391T missense mutation in ZIP8 results in reduced zinc transport, altered glutamate signaling, and an exacerbated immune and inflammatory response. Re-expression of wild-type ZIP8 or application of the zinc chelator ZX1 can rescue the observed phenotypes. These findings provide novel therapeutic targets relevant to schizophrenia.

Summary: Mutations in the LRRK2 gene lead to autosomal-dominant Parkinson's disease and may contribute to sporadic PD. Common genetic variations in LRRK2 could modify susceptibility to immunological disorders. The expression and phosphorylation of LRRK2 in human lymphoblasts suggest a potential role in immune and inflammatory responses.
