Expanding the scope of plant genome engineering with Cas12a orthologs and highly multiplexable editing systems

Yingxiao Zhang et al.

Summary: This study expands the targeting scope of Cas12a in crop genome engineering by screening Cas12a orthologs and developing a highly multiplexable editing system, with Mb2Cas12a showing outstanding editing efficiency and tolerance to low temperature. Additionally, two compact single transcript unit CRISPR-Cas12a interference systems were developed for multi-gene repression in rice and Arabidopsis.

CRISPAltRations: A validated cloud-based approach for interrogation of double-strand break repair mediated by CRISPR genome editing

Gavin Kurgan et al.

Summary: CRISPR systems enable targeted genome editing in various organisms through endogenous molecular pathways, and targeted genome resequencing can evaluate editing events. Building biological understanding into a novel analysis tool can improve results quality, and software validation shows superiority in annotating CRISPR-related events. Developing a user-friendly, cloud-hosted deployment allows non-bioinformaticians to use the software for resequencing analysis of CRISPR experiments.

Efficient Targeted Mutagenesis Mediated by CRISPR-Cas12a Ribonucleoprotein Complexes in Maize

Shujie Dong et al.

Summary: Recent advances in CRISPR-Cas genome editing technologies have enabled targeted mutagenesis and precise gene replacement in crop plants. Delivering Cas12a-gRNA ribonucleoprotein complex (RNP) directly to immature maize embryos through particle bombardment can efficiently generate genome edited lines. Co-delivering Cas12a RNP with a selectable marker gene cassette and selecting induced callus cultures with mannose can significantly increase gene editing efficiency.
