Membrane Damage during Ferroptosis Is Caused by Oxidation of Phospholipids Catalyzed by the Oxidoreductases POR and CYB5R1

Bo Yan et al.

Summary: Ferroptosis is a form of necrotic cell death caused by iron-dependent peroxidation of polyunsaturated phospholipids on cell membranes, which is carried out by oxidoreductases transferring electrons to oxygen to generate hydrogen peroxide for lipid peroxidation. This process needs to be counteracted by antioxidant systems to prevent cell death.

Ferroptosis: mechanisms, biology and role in disease

Xuejun Jiang et al.

Summary: Ferroptosis, as a form of regulated cell death driven by iron-dependent phospholipid peroxidation, has seen significant growth in research in recent years. Studies have focused on molecular mechanisms, regulation, and functions of ferroptosis, linking this cell death modality to various pathologies and proposing its roles in normal physiology and potential therapeutic targeting.

Peroxisome-driven ether-linked phospholipids biosynthesis is essential for ferroptosis

Weiwei Cui et al.

Summary: Ferroptosis is primarily induced by peroxidation of long-chain PUFA, but the role of SFA remains unclear. The study identified FAR1 as a key factor in SFA-mediated ferroptosis, involved in the synthesis of alkyl-ether lipids and plasmalogens, revealing a new pathway for ferroptosis.

Ferroptotic cell death triggered by conjugated linolenic acids is mediated by ACSL1

Alexander Beatty et al.

Summary: Inhibition of lipid peroxidase GPX4 promotes ferroptotic cell death. A complementary approach using conjugated linolenic fatty acids has been identified, triggering lipid peroxidation and ferroptosis via ACSL1, DGAT1/2, and neutral lipids. This may provide a potential therapeutic strategy for cancer treatment.

Non-enzymatic lipid peroxidation initiated by photodynamic therapy drives a distinct ferroptosis-like cell death pathway

Sufang Shui et al.

Summary: The study shows that photodynamic therapy (PDT) can induce lipid peroxidation and cell death similar to ferroptosis, termed as liperoptosis, which is independent of lipoxygenase and ACSL4. Using a clinically approved photosensitizer, PDT effectively inhibits tumor growth through triggering lipid peroxidation and cell death. These findings suggest the potential value of using PDT to induce lipid peroxidation and ferroptosis-like cell death in vivo for cancer treatment and exploring the immunological effects of PDT.

HIF-2α activation potentiates oxidative cell death in colorectal cancers by increasing cellular iron

Rashi Singhal et al.

Summary: This study reveals the essential role of HIF-2α in colorectal cancer (CRC) progression, demonstrating its regulation of cell death via modulation of cellular iron and oxidation. Inhibitors targeting HIF-2α can enhance cancer cells' susceptibility to oxidative cell death.
