Summary: The epidermis is a stratified epithelium with polarized structural and functional features across multiple cell layers. Desmosomes and their associated intermediate filaments play a key role in mechanical polarization in the epidermis, regulating different forces experienced by basal and suprabasal cells for layer-specific functions. Disengaging desmosomes from intermediate filaments accelerates differentiation markers expression and activates signaling pathways that contribute to tight junctions and barrier function maintenance in the epidermis.
