Thousands of high-quality sequencing samples fail to show meaningful correlation between 5S and 45S ribosomal DNA arrays in humans

Ashley N. Hall et al.

Summary: The study investigated copy number variation of ribosomal RNA genes (rDNA) and found no differences in probands with autism spectrum disorder and unaffected siblings. Validation results suggest that the previously reported concerted copy number variation between 45S and 5S arrays may be due to variable data quality, and meaningful correlation between 45S and 5S copy numbers was not detected in various datasets.

Methylation of 45S Ribosomal DNA (rDNA) Is Associated with Cancer and Aging in Humans

Fengqing Shao et al.

Summary: The study found that rDNA methylation in most cancer cases exhibited a consistent pattern, with hypermethylation in the coding region and hypomethylation in the promoter region, likely facilitating cancer cell proliferation and metastasis. In contrast, both the coding and promoter regions of rDNA showed increasing methylation during aging, suggesting a decline in rDNA activity. The discovery of rDNA methylation also hints at its potential application as an epigenetic biomarker in the diagnosis of cancer and aging.

Erosion of human X chromosome inactivation causes major remodeling of the iPSC proteome

Alejandro J. Brenes et al.

Summary: X chromosome inactivation (XCI) leads to a remodeling of the proteome in human induced pluripotent stem cells (iPSCs), with increased RNA and protein expression from X-linked genes. XCI erosion affects both X-linked and autosomal genes' protein expression, ultimately impacting a wider range of proteins and disease-linked loci.

The human ribosomal DNA array is composed of highly homogenized tandem clusters

Yutaro Hori et al.

Summary: Recent studies have revealed that human cells have a relatively regular rDNA structure, with variations in the noncoding regions of each rDNA copy. Nanopore sequencing further showed that all noncoding regions are heavily methylated, while about half of the coding regions are clearly unmethylated.
