Treatment strategies for cryptococcal infection: challenges, advances and future outlook

Summary: Cryptococcus species, particularly Cryptococcus neoformans and Cryptococcus gattii, have a significant impact on global human health, with almost a quarter of a million cases and 181,000 deaths from cryptococcal meningitis annually. Despite limited treatment options and high mortality rates, only three major classes of drugs are approved for clinical use. Challenges in developing new therapies include host toxicity, pathogen resistance, and the limited efficacy of existing antifungal agents. Research efforts are focused on understanding resistance mechanisms, exploring novel therapeutic strategies, and accelerating drug discovery through genomics technologies and high-throughput screening.
