Salidroside alleviates diabetic neuropathic pain through regulation of the AMPK-NLRP3 inflammasome axis

Summary: This study found that salidroside alleviates diabetic neuropathic pain by activating AMPK, inhibiting NLRP3 inflammasome activation, improving insulin resistance, and relieving oxidative stress. In vitro experiments also showed that salidroside regulated the AMPK-NLRP3 inflammasome axis in HG-treated DRGs. AMPK inhibition in vivo or silencing in vitro abolished the beneficial effects of salidroside on diabetic neuropathic pain.

The NLRP3-related inflammasome modulates pain behavior in a rat model of trigeminal neuropathic pain

Summary: In this study, it was found that in a rat model of trigeminal neuropathic pain, the level of NLRP3-related inflammasomes was upregulated in microglia after nerve damage. The inhibition of early expression of NLRP3 significantly reduced proinflammatory cytokines expression and alleviated mechanical allodynia. This suggests that targeting NLRP3 may be a potential therapeutic approach for neuropathic pain.

Additionally, salidroside may exert its protective effects through reducing oxidative stress levels, inhibiting p21 and p16 protein expressions, and promoting telomerase reverse transcriptase protein expression.
