Cell spheroids as a versatile research platform: formation mechanisms, high throughput production, characterization and applications

Monize Caiado Decarli et al.

Summary: Three-dimensional (3D) cell culture, especially using spheroids, is a powerful tool for mimicking the in vivo structure and microenvironment of healthy tissues and solid tumors. Engineering processes to obtain uniform 3D cell spheroids, considering mass transfer and shear stress, as well as computational and mathematical modeling, are important for developing large tissues and complex organs. Challenges in analyzing morphological parameters, cell quantification, viability, gene expression profiles, metabolic behavior, and high-content analysis need to be addressed to maximize the potential applications of spheroids in various research fields.

Kinship of conditionally immortalized cells derived from fetal bone to human bone-derived mesenchymal stroma cells

S. Marozin et al.

Summary: The hFOB 1.19 cell line is proposed as an accessible experimental model for studying osteoblast biology in relation to drug development and biomaterial engineering. hFOB cells show characteristics reminiscent of bmMSC, such as colony formation, migration capacity, and the propensity to grow as multicellular aggregates. However, after prolonged culture, hFOB cells acquired delayed growth rate and displayed morphological abnormalities similar to bmMSC, with decreased proliferation rates and increased expression of cell cycle inhibitors.
