Summary: This study found that p43 (-/-) mice did not show obvious hearing loss in juvenile stages, but developed premature and more severe age-related hearing loss due to loss of cochlear sensory cells and degeneration of spiral ganglion neurons. The exacerbated age-related hearing loss in p43 (-/-) mice was associated with early decrease in SIRT1 expression, imbalance in pro-apoptotic and anti-apoptotic proteins, mitochondrial dysfunction, oxidative stress, and inflammation. Additionally, p43 (-/-) mice were more susceptible to noise-induced hearing loss.

Summary: The iodide recycling enzyme, iodotyrosine deiodinase (IYD), is an understudied molecular mechanism through which environmental chemicals can potentially cause thyroid disruption. A screening assay was developed and used to identify 194 IYD inhibitors, with 154 chemicals further tested for potency. This study expands the scope of thyroid-relevant molecular targets for chemical screening and aids in prioritizing chemicals for targeted in vivo testing for thyroid-related adverse outcomes.

Monocarboxylate transporter 8 deficiency: update on clinical characteristics and treatment

Summary: Deficiency in the thyroid hormone transporter MCT8 leads to defective thyroid hormone transport, causing severe impact on neurodevelopment and chronic thyrotoxic state in peripheral tissues. Patients with MCT8 deficiency have severely impaired life expectancy, with increased mortality associated with lack of head control and being underweight at young age. Treatment with the thyroid hormone analogue triiodothyroacetic acid appears to be a promising therapy for this rare disorder.
