Hybrid Chemo-, Bio-, and Electrocatalysis for Atom-Efficient Deuteration of Cofactors in Heavy Water

Jack S. Rowbotham et al.

Summary: The study explores a range of catalyst systems for generating deuterium-labeled nicotinamide cofactors and considers their application in driving reductive deuteration reactions. It is shown that the inclusion of an electronically coupled NAD(+)-reducing enzyme offers significant advantages over purely metal systems, yielding exclusively [4S-H-2]-NADH.

Enantioselective decarboxylative protonation and deuteration of β-ketocarboxylic acids

Haruna Mizutani et al.

Summary: Enantioselective decarboxylative protonation of tetralone-derived beta-ketocarboxylic acids was achieved with up to 89% enantiomeric excess (ee) in the presence of a chiral primary amine catalyst. Furthermore, this method was applied to enantioselective deuteration to afford the corresponding alpha-deuterioketones with up to 88% ee.
