Enantioselective Vinylogous Mannich Reaction of Acyclic Vinylketene Silyl Acetals with Acyclic Ketimines

Kazuki Ogura et al.

Summary: A novel enantioselective vinylogous Mannich reaction was developed for the synthesis of acyclic delta-amino-alpha,beta-unsaturated carbonyl compounds, achieving excellent yields and enantioselectivities using a specific catalyst. This study provides an efficient route for the synthesis of compounds in this specific category.

Regio- and Stereoselective Cascade of β,γ-Unsaturated Ketones by Dipeptided Phosphonium Salt Catalysis: Stereospecific Construction of Dihydrofuro-Fused [2,3-b] Skeletons

Hongkui Zhang et al.

Summary: Chiral (dihydro)furo-fused heterocycles are important structural elements in various natural products, functional materials, and pharmaceuticals. An efficient method using dipeptide-phosphonium salt catalysis has been developed to prepare structurally fused heterocyclic compounds with high yields and excellent stereoselectivities. Mechanistic studies revealed that the bifunctional phosphonium salt controlled the regio- and stereoselectivities of the reaction, particularly through initial ketone alpha-addition and hydrogen-bonding interactions.

Organocatalytic Asymmetric Dearomatization Reaction for the Synthesis of Axial Chiral Allene-Derived Naphthalenones Bearing Quaternary Stereocenters

Alemayehu Gashaw Woldegiorgis et al.

Summary: The highly regio-, diastereo-, and enantioselective dearomatization reaction of 1-substituted 2-naphthols and beta,gamma-alkynyl-alpha-imino esters with complete atom economy is disclosed via chiral phosphoric acid catalysis. This protocol provides facile and efficient access to asymmetric construction of axially chiral allene-derived naphthalenones bearing quaternary stereocenters in good yields with high diastereoselectivities and enantioselectivities.

Asymmetric catalytic alkynylation of thiazolones and azlactones for synthesis of quaternary α-amino acid precursors

Beibei Meng et al.

Summary: Asymmetric alkynylation of thiazolones and azlactones with alkynylbenziodoxolones as electrophilic alkyne sources catalyzed by thiourea phosphonium salt provides the desired products in good yields and enantioselectivities. Moreover, ring-opening of the alkynylation products efficiently leads to alpha,alpha-disubstituted alpha-amino acid derivatives without loss of enantioselectivity.

Construction of chiral α-tert-amine scaffolds via amine-catalyzed asymmetric Mannich reactions of alkyl-substituted ketimines

Chihiro Homma et al.

Summary: This study demonstrated a highly diastereodivergent synthesis of beta-amino aldehydes with a chiral alpha-tert-amine moiety, utilizing less-activated Z-ketimines in amine-catalyzed Mannich reactions with aldehydes. This method overcame the limitations of structural variations in ketimines, achieving high diastereoselectivity.
