Base-Promoted Cascade Reactions for the Synthesis of 3,3-Dialkylated Isoindolin-1-ones and 3-Methyleneisoindolin-1-ones

Antonio Macchia et al.

Summary: Cascade reactions of ortho-carbonyl-substituted benzonitriles with ((chloromethyl)sulfonyl)benzenes as pronucleophiles can lead to new isoindolin-1-ones with a tetrasubstituted C-3 position or (Z)-3-(sulfonyl-methylene)isoindolin-1-ones. These reactions are carried out under mild conditions without the need for metal catalysis, promoted only by environmentally benign K2CO3, showing potential applications for the synthesis of aristolactam natural products.

Metal-Free Cascade Formation of Intermolecular C-N Bonds Accessing Substituted Isoindolinones under Cathodic Reduction

Zirong Zou et al.

Summary: An electrochemical protocol has been developed for the construction of substituted isoindolinones via reduction/amidation of 2-carboxybenzaldehydes and amines. This reaction occurs under metal-free and external-reductant-free electrolytic conditions, forming intermolecular C-N bonds and providing isoindolinones in moderate to good yields. The deuterium-labeling experiment demonstrates that hydrogen in the product is mainly derived from H2O in the system.

Ligand Enabled Pd(II)-Catalyzed γ-C(sp3)-H Lactamization of Native Amides

Shuang Liu et al.

Summary: A novel catalytic system was reported for the intramolecular gamma-C-H amination of aliphatic amides derived from amino acids, enabling the convenient synthesis of lactams, isoindolinones, and 2-imidazolidinones. This protocol demonstrates the utility of utilizing N-acyl amino acids as directing groups and cyclization partners, tert-butyl hydrogen peroxide as the sole oxidant, and a broad substrate scope for the construction of important structural cores of medicinal compounds.

Tao Ma et al.

Summary: A convenient and environmentally friendly synthetic route has been developed for the transformation from 2-vinylbenzimide to 3-hydroxy-isoindolinones, promoted by visible light via iron/disulfide catalysis and molecular oxygen oxidation. A variety of 3-hydroxy-isoindolinones were obtained in moderate to good yields, showing excellent functional group compatibility and broad substrate scope. Further mechanistic investigations revealed that dioxetane might act as a key intermediate in the reaction.

Alumina-Supported Alkali and Alkaline Earth Metal-Based Catalyst for Selective Decarboxylation of Itaconic Acid to Methacrylic Acid

Hadi Ali et al.

Summary: In this study, selective decarboxylation of itaconic acid to methacrylic acid using different metals supported on Al2O3 was investigated. Among the metals, calcium proved to be the most effective catalyst, achieving the highest yield of MAA and also producing a significant amount of alpha-hydroxyisobutyric acid as a byproduct. This can be attributed to the higher number of basic sites present in the 5 wt% Ca/Al2O3 catalyst.

Pd-Catalyzed C(sp2)-H olefination: synthesis of N-alkylated isoindolinone scaffolds from aryl amides of amino acid esters

Manish K. Gupta et al.

Summary: The paper describes the synthesis of N-alkyl-3-methenyl chiral isoindolinone derivatives from aryl amides of l-amino acids and non-activated alkene via Pd-catalyzed C(sp(2))-H olefination. The amino acid residue acts as a directing group for olefination at the aryl ring and cyclization occurs at the amide NH. This methodology could be useful in transforming standard amino acids into respective chiral isoindolinone derivatives.
