HILIC-UPLC-MS for high throughput and isomericN-glycan separation and characterization in Congenital Disorders Glycosylation and human diseases

Angela Messina et al.

Summary: N-glycan analysis can uncover disease-associated biomarkers and diagnose genetic disorders, and liquid chromatography-mass spectrometry is a powerful analytical method. Using the RapiFluor-MS method for N-glycan identification and separation allows for detailed characterization of structural details in patients' serum, differentiation between structural isomers and isomers with different linkage types.

Glycoform-resolved pharmacokinetic studies in a rat model employing glycoengineered variants of a therapeutic monoclonal antibody

David Falck et al.

Summary: Good pharmacokinetic behavior is crucial for the efficacy of therapeutic monoclonal antibodies. Fc glycosylation plays a key role in stability and effector functions of mAbs, but the effects of different IgG Fc glycoforms on antibody PK are not well understood. Glycoengineering and glycoform-resolved PK measurements showed increased clearance for high-mannose and hybrid-type glycoforms, while sialylation did not affect clearance as expected.
