First Small-Molecule Inhibitors Targeting the RNA-Binding Protein IGF2BP2/IMP2 for Cancer Therapy

Summary: This study validated IMP2 as a potential target for colorectal cancer and established a screening assay for small-molecule inhibitors of IMP2. In vitro and in vivo studies demonstrated that IMP2 knockout reduces tumor cell proliferation and growth. Ten compounds from different libraries showed target specificity and were validated in vitro. The three most active compounds also exhibited reduced tumor xenograft growth in vivo. These findings support IMP2 as a druggable target to reduce tumor cell proliferation.

Synthesis and evaluation of dual-action kanglemycin-fluoroquinolone hybrid antibiotics

Hybrid antibiotics, composed of two covalently linked antibiotic entities, offer a strategy to overcome resistance. This study evaluates a series of semi-synthetic hybrid antibiotics formed by linking Kang A, a rifampicin analog, with fluoroquinolones. The results suggest that these hybrids may provide new avenues for generating antibiotics effective against drug-resistant infections.

Total synthesis and chemical stability of pseudouridimycin

Summary: We report the chemical synthesis of pseudouridimycin (1), an antimicrobial natural product that selectively inhibits bacterial RNA polymerase. Intramolecular hydroxamate bond scission was found to be a major decomposition pathway for 1 in aqueous buffer. Replacement of the hydroxamate bond with a tertiary amide resulted in a conformational isostere resistant to degradation. These findings lay the foundation for the design and synthesis of analogues with improved chemical stability and biological activity.
