Nanocatalytic Theranostics with Glutathione Depletion and Enhanced Reactive Oxygen Species Generation for Efficient Cancer Therapy

Lian-Hua Fu et al.

Summary: Chemodynamic therapy (CDT) is a promising method for killing cancer cells by converting intracellular hydrogen peroxide (H2O2) into highly toxic hydroxyl radicals ((OH)-O-center dot). An intelligent nanocatalytic theranostics, PGC-DOX, was developed to address the limitations of endogenous H2O2 and high GSH levels in tumor cells, leading to efficient cancer therapy. The system combines H2O2 self-supply, GSH-elimination, and DOX-induced chemotherapy to effectively inhibit tumor growth with minimal side effects in vivo.

Immunometabolism in the Tumor Microenvironment

Dominic G. Roy et al.

Summary: Advances in immunotherapy have highlighted the importance of antitumor immune responses in controlling cancer, but the tumor microenvironment poses obstacles to immune cell function through metabolic and immunosuppressive mechanisms. Studies have shown the characteristic metabolic reprogramming of different immune cell subsets, the shaping of immune cell metabolism and function by TME, and the potential of therapeutic interventions to enhance antitumor immunity by improving immune cell metabolic fitness and alleviating metabolic constraints in the TME.
