In-vitro and In-vivo Photocatalytic Cancer Therapy with Biocompatible Iridium(III) Photocatalysts

Summary: Ir3 is an Ir-III photocatalyst with strong light absorption, high biocompatibility, and excellent photocatalytic anticancer properties. It showed selective photo-cytotoxicity against drug-resistant cells while remaining dormant to normal cells, and exhibited efficient photo-catalytic oxidation of cellular components leading to necrosis and apoptosis of cancer cells. Ir3 also demonstrated high biocompatibility and anticancer efficiency in in vivo models.

Anticancer effect evaluation in vitro and in vivo of iridium(III) polypyridyl complexes targeting DNA and mitochondria

Summary: The investigation revealed that the iridium complexes Ir1-3 exhibit high cytotoxicity to most tumor cells, especially SGC-7901 cells. These complexes can increase intracellular reactive oxygen species levels, reduce mitochondrial membrane potential, induce apoptosis, and inhibit cell proliferation. The experimental and theoretical findings indicate that Ir1-3 induce apoptosis in SGC-7901 cells through mitochondrial dysfunction and DNA damage pathways, thus exerting anti-tumor activity in vitro and in vivo.

Summary: The limited therapeutic efficacy of photodynamic therapy (PDT) to hypoxic solid tumors has been improved by two new benzothiophenylisoquinoline-derived cyclometalated Ir-III complexes, IrL1 and MitoIrL2. Both complexes demonstrated a type I PDT process and induced photoinduced ferroptosis in tumor cells under hypoxia, with MitoIrL2 outperforming IrL1 in inhibiting tumor growth through a synergistic effect of ferroptosis and apoptosis.

Co-expression of Pol β and PARG in human tumors is associated with poor patient survival, suggesting potential therapeutic strategies targeting Pol β in ovarian cancers.

An Iridium Complex as an AIE-active Photosensitizer for Image-guided Photodynamic Therapy
