mTOR-mediated autophagy in the hippocampus is involved in perioperative neurocognitive disorders in diabetic rats

Xiaohui Chen et al.

Summary: The study showed that pretreatment with rapamycin can inhibit mTOR hyperactivation, restore autophagic function, effectively reduce tau hyperphosphorylation, A beta deposition, and apoptosis in the hippocampus in diabetic rats with PND. Furthermore, surgical trauma-induced neurocognitive disorders in diabetic rats can also be reversed by pretreatment with rapamycin.

Varenicline improved laparotomy-induced cognitive impairment by restoring mitophagy in aged mice

Jianan Wang et al.

Summary: This study found that laparotomy in aged mice caused persistent cognitive impairment and anxiety. This impairment may be associated with mitochondrial dysfunction, oxidative stress, and dysregulated autophagy. The study also found that chronic varenicline administration improved cognitive impairment in these mice.

Inflammatory monocytes and microglia play independent roles in inflammatory ictogenesis

Charles L. Howe et al.

Summary: This study investigated the relative contribution of inflammatory monocytes and microglia in ictogenesis using the Theiler's murine encephalomyelitis virus (TMEV) model. The results showed that the microglial response did not scale with the inoculum, while the inflammatory monocyte response scaled with the amount of infectious virus. Inoculation conditions that drove inflammatory monocyte infiltration resulted in severe seizures and EEG abnormalities, while low-level inoculation conditions associated with microglial activation resulted in milder seizures and EEG phenotypes.

Biomarkers of Cerebral Injury for Prediction of Postoperative Cognitive Dysfunction in Patients Undergoing Cardiac Surgery

Sebastian Wiberg et al.

Summary: The study evaluated the ability of biomarkers NSE, tau, NFL, and GFAP to predict postoperative cognitive dysfunction (POCD) in cardiac surgery patients. Elevated levels of tau and GFAP were found in patients with POCD at discharge, but the biomarkers only had moderate predictive abilities for POCD. Postoperative levels of NSE were not associated with POCD at discharge.

Estrogen Attenuates Traumatic Brain Injury by Inhibiting the Activation of Microglia and Astrocyte-Mediated Neuroinflammatory Responses

Jin Wang et al.

Summary: The study demonstrated that E2 treatment significantly alleviated TBI-induced neuroinflammatory injury by inhibiting microglia and astrocyte activation, reducing neurotoxic astrocyte activation, and inhibiting the TLR4/NF-kappa B pathway. E2 treatment may be a potential therapy strategy for TBI-induced neuroinflammation injury.

miRNA-124-3p targeting of LPIN1 attenuates inflammation and apoptosis in aged male rats cardiopulmonary bypass model of perioperative neurocognitive disorders

Jiang Han et al.

Summary: The study found that miR-124-3p was significantly reduced in a PND rat model, and its overexpression could alleviate inflammation, cell apoptosis, and improve cognitive function. It was suggested that miR-124-3p may improve the state of PND by modulating LPIN1, providing a new strategy for preventing and treating PND.

Abnormal phosphorylation of tau protein and neuroinflammation induced by laparotomy in an animal model of postoperative delirium

Le Yu et al.

Summary: Postoperative delirium is an acute neuropsychological disturbance that may be influenced by neuroinflammation and abnormal tau phosphorylation. In a study with mice undergoing laparotomy, significant changes in inflammatory responses and tau protein levels were observed at 4 hours and 24 hours postoperatively, indicating a potential link between these factors and cognitive dysfunction.

Postoperative cognitive dysfunction is made persistent with morphine treatment in aged rats

Stephanie M. Muscat et al.

Summary: Postoperative cognitive dysfunction (POCD) is a collection of cognitive impairments experienced by individuals following surgery, with persistent POCD most commonly affecting older individuals. In a study on aged rats, it was found that postsurgical treatment with morphine led to long-term memory impairments through a neuroinflammatory mechanism rather than pain modulation. Central blockade of IL-1b signaling completely prevented the memory impairment in rats, suggesting a potential target for future treatment discoveries.

Reversing Postcardiopulmonary Bypass Associated Cognitive Dysfunction Using k-Opioid Receptor Agonists to Regulate Microglial Polarization via the NLRP3/Caspase-1 Pathway

Pei Song et al.

Summary: This study investigated the effects of KOR agonists on neuroinflammation and microglia polarization post-cardiopulmonary bypass, finding that KORs can inhibit inflammation through the NLRP3/caspase-1 signaling pathway, alleviate hippocampal damage caused by CPB, and improve PND in rats.

Neuroinflammation: Integrated Nervous Tissue Response through Intercellular Interactions at the Whole System Scale

Daniele Nosi et al.

Summary: Different cell populations within the nervous tissue, such as microglia and astrocytes, play important roles in neurodegenerative diseases by triggering neuroinflammation. This chronic neuroinflammation is characterized by a shift in the activities of astrocytes and microglia from cytoprotective to cytotoxic. Overall, the interactions and activities of various cell types in the nervous system contribute to the complex process of neuroinflammation.

Neuroinflammation in Alzheimer's Disease: Focus on NLRP1 and NLRP3 Inflammasomes

Eliana C. de Brito Toscano et al.

Summary: Alzheimer's disease is the main cause of dementia globally, with the definitive diagnosis based on the identification of cerebral deposition of Amyloid beta plaques and neurofibrillary tangles. Research suggests that inflammasomes may play a significant role in AD, but there is a lack of studies evaluating their role in patients with AD.

IL-6 and IL-1β upregulation and tau protein phosphorylation in response to chronic alcohol exposure in the mouse hippocampus

Cihao Jiang et al.

Summary: Chronic alcohol exposure impairs spatial recognition ability in mice, upregulates the expression of IL-1 beta and IL-6 in serum and the hippocampus, increases phosphorylation of tau protein in the hippocampus, and downregulates PSD95 and synaptophysin protein levels. This suggests that hippocampal IL-1 beta, IL-6, and phosphorylated tau proteins may play a role in the neurotoxic mechanism of chronic alcohol exposure by mediating synaptic dysfunction.

Cong-mei Wang et al.

Summary: Sevoflurane is widely used in anesthesia for surgical patients, but may increase the incidence of postoperative cognitive dysfunction (POCD). POCD affects patients' lives and adds burden to patients and their families. Understanding the mechanism of Sevoflurane-induced POCD is crucial for prevention and treatment.
