The ingenol-based protein kinase C agonist GSK445A is a potent inducer of HIV and SIV RNA transcription

Afam A. Okoye et al.

Summary: The study discusses a novel latency-reversing agent called GSK445A, which has shown promise in inducing HIV and SIV latency reversal in vitro, and has demonstrated a tolerable safety profile in nonhuman primates. This suggests potential for further evaluation of this PKC-agonist in strategies to cure HIV.

Everolimus, an mTORC1/2 inhibitor, in ART-suppressed individuals who received solid organ transplantation: A prospective study

Timothy J. Henrich et al.

Summary: Pharmacologic inhibition of mTOR with everolimus in HIV-infected solid organ transplant recipients did not significantly affect overall HIV DNA and RNA levels, but participants with higher everolimus trough levels showed lower RNA levels after cessation of treatment. Time-averaged trough levels of everolimus correlated with greater inhibition of mTOR gene pathway transcriptional activity, and treatment also led to decreased PD-1 expression on certain T cell subsets.

Antigen-driven clonal selection shapes the persistence of HIV-1-infected CD4(+) T cells in vivo

Francesco R. Simonetti et al.

Summary: The study highlights that antigen-driven clonal selection plays a major role in HIV-1 persistence, posing a difficult challenge for eradication efforts. The research demonstrates a link between antigen responsiveness, proviral sequence, integration site, and TCR beta sequence in understanding the mechanisms behind maintaining the HIV-1 reservoir.

The emerging role of microRNA in regulating the mTOR signaling pathway in immune and inflammatory responses

Nazanin Nazari et al.

Summary: mTOR, a critical regulator of cellular events and immune function, interacts with miRNAs to modulate immune responses. miRNAs target the mTOR signaling pathway to regulate immune reactions. Dysregulation of miRNAs and the mTOR pathway may contribute to immune-related diseases.

CD8+ T cells fail to limit SIV reactivation following ART withdrawal until after viral amplification

Afam A. Okoye et al.

Summary: The study found that CD8+ T cell depletion did not significantly affect the control of SIV reactivation during ART, but after ART cessation, rhesus macaques with CD8+ T cell depletion showed a significant increase in post-ART plasma viremia.

Integration in oncogenes plays only a minor role in determining the in vivo distribution of HIV integration sites before or during suppressive antiretroviral therapy

John M. Coffin et al.

Summary: HIV-infected cells show a preference for integrating into highly-expressed genes, but this preference decreases over time. However, integration into specific regions of certain genes can promote cell growth and survival, despite being a minor factor in promoting the overall growth and survival of infected cells during ART.

Rapamycin Inhibits Glioma Cells Growth and Promotes Autophagy by miR-26a-5p/DAPK1 Axis

Zheng Wang et al.

Summary: The study revealed that rapamycin exerts anti-tumor effects in glioma cells by promoting autophagy, inhibiting cell proliferation, and promoting apoptosis. The activation of the miR-26a-5p/DAPK1 pathway by rapamycin played a crucial role in these processes.
