High-Z-Sensitized Radiotherapy Synergizes with the Intervention of the Pentose Phosphate Pathway for In Situ Tumor Vaccination

Summary: In situ tumor vaccination is achieved through the integration of Phy@PLGdH nanosheets, which enhances radiation therapy-induced immunogenic cell death. The nanosheets exhibit improved radiosensitization and tumor penetrability, while Phy-mediated PPP intervention amplifies the oxidative stress and DNA damage caused by PLGdH-sensitized radiation therapy. This results in effective immunogenic cell death and enhanced immunogenicity of irradiated tumor cells. Furthermore, Phy@PLGdH-sensitized radiation therapy successfully triggers a robust CD8(+)-T-cell-dependent antitumor immune response, which enhances checkpoint blockade immunotherapies against primary and metastatic tumors.
