Solid-Binding Peptide-Guided Spatially Directed Immobilization of Kinetically Matched Enzyme Cascades in Membrane Nanoreactors

Deniz T. Yucesoy et al.

Summary: This study demonstrates a modular cascade flow reactor using a solid-binding peptide-directed immobilization strategy on anodic aluminum oxide (AAO) monoliths for two-step biocatalytic production of L-lactate with concomitant regeneration of NADH. The strategy provides insight for controlling surface orientation, spatial localization, and loading of multiple enzymes on solid supports, enabling the construction of high-power cascadic devices.

Enzyme entrapment, biocatalyst immobilization without covalent attachment

Hasan T. Imam et al.

Summary: Entrapment immobilization involves trapping an active species, often a catalyst, within a material forming a solid or gel. It is used to immobilize enzymes and for whole cell biocatalysis. Despite its advantages of no formal interaction with the protein needed, entrapping isolated enzymes has lagged behind other methods like covalent attachment and crosslinking due to risks of enzyme leaching and mass transfer limitations. Key methods are characterized and recent innovations are highlighted.

Laccase aggregates via poly-lysine-supported immobilization onto PEGA resin, with efficient activity and high operational stability and can be used to degrade endocrine-disrupting chemicals

Hiroshi Yamaguchi et al.

Summary: Utilizing a simple and cost-effective immobilization technique, laccase was immobilized onto PEGA resin as Lac-PEGA, demonstrating similar thermal and pH stabilities to free laccase. Lac-PEGA showed improved stability in organic solvents, could be reused for 50 cycles, and effectively removed BPA in batch and flow reactors with high efficiency, making it a promising catalyst for various applications in chemistry and biotechnology.
