New insight into the codon usage and medium optimization toward stable and high-level 5-aminolevulinic acid production in Escherichia coli

Tzu-Hsuan Yu et al.

Summary: The study demonstrated that codon adaptation index and tRNA adaptation index influence the expression level and folding of recombinant ALAS gene in E. coli. Genomic integration and co-expression of chaperone helped increase 5-ALA production in E. coli. Addition of ferric ion during continuous culture boosted cell growth and 5-ALA production.

Biotechnological advances for improving natural pigment production: a state-of-the-art review

Xiaomei Lyu et al.

Summary: Due to the increase of people's health awareness and the discovery of novel pharmacological effects, natural pigments are experiencing a rapidly growing global market. However, traditional production methods are limited by the source and low content of target pigment compounds. To achieve industrial-scale production, many efforts have been made to increase pigment production through the development of in vitro plant cell/tissue culture systems and optimization of microbial cultivation methods. Synthetic biology has also provided new approaches for the biosynthesis of pigments from different sources. This review comprehensively summarizes the innovative methods and strategies for optimizing and engineering both native and heterologous producers of natural pigments, presents current progress in the production of several representative high-value natural pigments, and discusses the remaining challenges and future perspectives.

Low-Carbon-Footprint Production of High-End 5-Aminolevulinic Acid via Integrative Strain Engineering and RuBisCo-Equipped Escherichia coli

Ying-Chen Yi et al.

Summary: This study focused on improving the expression and activity of ALA synthase (ALAS), utilizing the pdxY gene for cofactor regeneration, and introducing RuBisCo and Prk to reduce CO2 emissions and enhance ALA production. By implementing these strategies, a higher ALA yield was achieved for application in antibacterial photodynamic therapy.

Inactivation kinetics of Bacillus cereus vegetative cells and spores from different sources by antimicrobial photodynamic treatment (aPDT)

Leonardo do Prado-Silva et al.

Summary: Antimicrobial photodynamic treatment (aPDT) using new methylene blue (NMB) as photosensitizer (PS) with red light effectively inactivated Bacillus cereus vegetative cells and spores. Different strains of B. cereus showed varying susceptibility to aPDT, with strain ATCC 14579 being the most susceptible at a relatively low fluence. The Weibull model provided a good fit for inactivation data, estimating kinetic parameters for microbial inactivation.
