Aldehyde-methacrylate-hyaluronan profited hydrogel system integrating aligned and viscoelastic cues for neurogenesis

Summary: In this study, we developed a hydrogel system with orthogonal viscoelasticity and nano-topography by modifying hyaluronan. The matrix nanoorientation and viscoelasticity were found to have positive effects on neurites outgrowth, neural cell elongation, and neuronal differentiation. Moreover, implantation of the viscoelastic and nano-aligned hydrogels showed enhanced repair of spinal cord injury, including improved pathological microenvironment, facilitated endogenous neurogenesis, axon regeneration, and motor function restoration.

Nanoengineered on-demand drug delivery system improves efficacy of pharmacotherapy for epilepsy

Summary: This study reports a nanoengineered drug delivery system for improved delivery efficiency and on-demand drug release of antiepileptic drugs (AEDs). The system utilizes receptor-mediated transcytosis and blood-brain barrier disruption induced by photothermal conversion to achieve brain-targeted delivery. The system has been shown to effectively inhibit seizures upon epileptiform abnormalities, using only one-fifth of the conventional dosage, and holds promise for clinical applications.

Antioxidative and Conductive Nanoparticles-Embedded Cell Niche for Neural Differentiation and Spinal Cord Injury Repair

Summary: This study developed difunctional polypyrrole nanoparticles incorporated into bioactive collagen/hyaluronan hydrogel to construct an antioxidative and conductive microenvironment for nerve regeneration. The embedded PPy protected BMSCs from oxidative damage and promoted neuronal differentiation with the synergy between conductivity and electrical stimulation, associated with PI3K/Akt and MAPK pathway. In SCI rats, the difunctional hydrogel laden with BMSCs restored bioelectric signal transmission, inhibited secondary damage, and facilitated neurogenesis, resulting in prominent nerve regeneration and functional recovery.
