Increased demand for NAD+ relative to ATP drives aerobic glycolysis

Alba Luengo et al.

Summary: Increasing the activity of the pyruvate dehydrogenase complex impairs cell proliferation by reducing the NAD(+)/NADH ratio, which is caused by increased mitochondrial membrane potential. When demand for NAD(+) exceeds the demand for ATP in cells, aerobic glycolysis is promoted despite available oxygen.

Type I collagen deletion in αSMA+ myofibroblasts augments immune suppression and accelerates progression of pancreatic cancer

Yang Chen et al.

Summary: The stromal desmoplastic reaction in pancreatic ductal adenocarcinoma involves significant accumulation of type I collagen (Col1). Deletion of Col1 specifically in myofibroblasts accelerates PDAC progression by increasing Cxcl5 expression in cancer cells and promoting recruitment of myeloid-derived suppressor cells. Targeting CXCR2 and CCR2 may help restrain accelerated PDAC progression in the absence of stromal Col1.

Fibroblast pyruvate carboxylase is required for collagen production in the tumour microenvironment

Summary: The abnormal collagen production by fibroblasts can influence cancer progression, with TCA cycle anaplerosis determining collagen production through pyruvate carboxylase, which could be a potential target to inhibit tumor desmoplasia.

Mitochondrial NADP+ is essential for proline biosynthesis during cell growth

Diem H. Tran et al.

Summary: Research has shown that mitochondrial NADPH is crucial for proline biosynthesis in cells, with NAD kinase 2 (NADK2) activity being necessary for the production of mitochondrial NADP(+). Deficiency in mitochondrial NADPH leads to the inability of cells to synthesize proline, impacting nucleotide and protein synthesis, thereby impeding cell growth.
