Summary: This study investigated the subsequent reaction pathways of diradical intermediates formed from acyclic enediynes based on maleimide-assisted rearrangement and cycloaromatization. Experimental results suggested that intramolecular HAT processes compete with intermolecular H-abstraction, indicating that DNA-cleavage ability of enediynes can be enhanced by halting HAT processes.

Cancer statistics for the year 2020: An overview

It estimated that there were nearly 19.3 million new cancer cases and almost 10 million cancer deaths worldwide in 2020, with the most commonly diagnosed cancers being female breast cancer and lung cancer, and the most common causes of cancer death being lung cancer and liver cancer.

Synthesis of maleimide-based enediynes with cyclopropane moieties for enhanced cytotoxicity under normoxic and hypoxic conditions

Summary: Maleimide-based enediynes with cyclopropane moieties were synthesized to enhance cytotoxicity towards tumor cells by rearranging diradical intermediates with longer separation and weaker interactions. These compounds exhibited similar cytotoxicity under hypoxic conditions, suggesting potential use in treating solid tumors with compromised drug efficiency. Docking studies indicated that diradical intermediates can induce DNA damage and cell death through an apoptosis pathway.
