Metal-free photosensitized oxyimination of unactivated alkenes with bifunctional oxime carbonates

Tuhin Patra et al.

Summary: A metal-free photosensitization protocol has been developed for the installation of both amine and alcohol functionalities into alkene feedstocks in a single step, demonstrating better regioselectivity compared to traditional methods. The use of orthogonal protection for amine and alcohol functionalities allows for the direct synthetic diversification of one functional handle without influencing the other. This process, utilizing readily available starting materials, offers a promising approach for various synthetic endeavors due to its convergent synthesis and mild reaction conditions.

Desymmetrization of Prochiral Cyclobutanones via Nitrogen Insertion: A Concise Route to Chiral γ-Lactams

Jan Sietmann et al.

Summary: Asymmetric access to gamma-lactams was achieved using (1S,2R)-1-amino-2-indanol for chiral induction, with downstream stereoinduction up to 88:12 dr. Mechanistic analysis revealed a Curtin-Hammett scenario, supported by spectroscopic, crystallographic, and computational studies. The method allows for the synthesis of optically pure gamma-lactams, including those with all-carbon quaternary stereocenters, and is exemplified by the formal synthesis of drug molecules baclofen, brivaracetam, and pregabalin.

Photocatalytic Anti-Markovnikov Radical Hydro- and Aminooxygenation of Unactivated Alkenes Tuned by Ketoxime Carbonates

Sheng-Qiang Lai et al.

Summary: A tunable photocatalytic method using ketoxime carbonates as reagents has been developed for anti-Markovnikov hydro- and aminooxygenation of unactivated alkenes. The reaction is initiated by N-O bond homolysis of ketoxime carbonates under visible-light photocatalysis, followed by generation of alkoxylcarbonyloxyl and iminyl radicals leading to carbon radical addition to alkenes. The carbon radical can either abstract a hydrogen atom to form hydrooxygenation product or be trapped by iminyl radical to yield aminooxygenation product.
