Optimization of a PDMS-Based Cell Culture Substrate for High-Density Human-Induced Pluripotent Stem Cell Adhesion and Long-Term Differentiation into Cardiomyocytes under a Xeno-Free Condition

Summary: Despite the hydrophobic surface of PDMS-based substrates limiting cell attachment and growth, this study found that a PDMS substrate coated with a combination of PDA and LME8 fragments can support the attachment and growth of hiPSCs, as well as their differentiation into iCMs. Compared to previous studies, the density of hiPSCs and their adhesion time on the PDMS surface were increased during iCM production. Additionally, this coated substrate required less manipulation and could be used under animal-free conditions. The results suggest that the PDA + LME8-coated PDMS substrate is a promising tool for cardiomyocyte production, tissue engineering, microfluidics, and organ-on-chip platforms.

P75 and S100 gene expression induced by cell-imprinted substrate and beta-carotene to nerve tissue engineering

Summary: In this study, adipose derived stem cells (ADSCs) were successfully differentiated into Schwann cells (SCs) using imprinting method and beta-carotene. The topography of SCs was imprinted on PDMS substrates, inducing differentiation signals in MSCs and upregulation of SC specific markers with the addition of beta-carotene. This study demonstrated the ability of Schwann cell imprinted substrates and beta-carotene to mimic SC morphology and induce neural differentiation in MSCs, without the need for expensive or unsafe chemical growth factors.

An integrated microfluidic device for stem cell differentiation based on cell-imprinted substrate designed for cartilage regeneration in a rabbit model

Summary: This study demonstrates the potential of the imprinting method in inducing chondrogenicity in stem cells, with successful regeneration of articular cartilage defects in rabbits after transplantation. The efficiency of the imprinting method was increased using microfluidic chip design and fabrication, leading to the precise placement of stem cells on chondrocyte patterns on the substrate.

Molecular Imprinting Strategies for Tissue Engineering Applications: A Review

Summary: Tissue Engineering (TE) is a promising approach to restore tissue damage, using biomaterials and growth factors to create scaffolding structures supporting cell growth. One current challenge is to create engineered constructs that mimic the complex microenvironment of natural tissue to promote cell migration, proliferation, and differentiation. Introducing molecularly imprinted polymers (MIPs) into scaffolds holds promise to enhance scaffold-cell interaction in TE applications, with potential for biophysical cues and drug delivery.

Donor-controlled clinical practice is suggested to minimize potential harm in MSC-based therapies.

The review also highlights key mechanobiological findings in innate immune cells that underpin our understanding of their role in the response to implantable biomaterials.
