Targeting cancer metabolism in the era of precision oncology

Zachary E. Stine et al.

Summary: In the past century, there have been significant advancements in targeting cancer metabolism, but progress in the past decade has been limited. Some metabolism-based drugs for cancer have been successfully developed and are in clinical trials. However, consideration of the metabolism of non-cancer cells is crucial for successful treatment of cancer.

Acetyl-CoA Synthetase 2: A Critical Linkage in Obesity-Induced Tumorigenesis in Myeloma

Zongwei Li et al.

Summary: The study indicates that acetyl-CoA synthetase 2 (ACSS2) plays an important role in obesity-related myeloma progression, interacting with oncoprotein interferon regulatory factor 4 (IRF4) to enhance tumor growth. Overexpression of ACSS2 contributes to IRF4 stability and gene transcription, reducing myeloma growth in an obese mouse model.

Hypoxia-Inducible Factor (HIF): Fuel for Cancer Progression

Saurabh Satija et al.

Summary: HIF-1 is an important pathway in the tumor microenvironment that regulates cellular responses to hypoxia by activating adaptive reactions through the transcription factor HIF-1 alpha. The dysregulation of HIF-1 alpha expression can lead to the loss of tumor suppressant activity in cells, influencing cancer progression. Recent attention by translational researchers has focused on understanding the crucial role of HIF in cancer and its potential implications for other diseases.

Cadmium promotes breast cancer cell proliferation, migration and invasion by inhibiting ACSS2/ATG5-mediated autophagy

Yidan Liang et al.

Summary: This study revealed that Cd exposure accelerates proliferation, migration, and invasion of MCF-7 and T47-D cells by suppressing ATG5-dependent autophagosome formation. Impairment of the ACSS2/ATG5-dependent autophagic flux promotes BC cell proliferation and metastasis stimulated by Cd.

Acetate stimulates lipogenesis via AMPKα signaling in rabbit adipose-derived stem cells

Lei Liu et al.

Summary: In this study, it was found that acetate treatment increased lipid deposition in rabbit ADSCs, along with higher levels of phosphorylated AMPK alpha and p38 MAPK proteins. Blocking AMPK alpha signaling reduced acetate-induced lipid accumulation, but p38 MAPK signaling was not affected. These findings suggest that AMPK alpha signaling pathway is associated with acetate-induced lipogenesis.

Acetate supplementation produces antidepressant-like effect via enhanced histone acetylation

Weibin Huang et al.

Summary: Acetate supplementation was found to significantly improve depression-like behaviors in mice and enhance dendritic branches and spine density of CA1 pyramidal neurons. Administration of acetate rescued down-regulated BDNF and TrkB levels. Chronic acetate treatment led to decreased transcription levels of HDAC2, HDAC5, HDAC7, HDAC8, increased transcription levels of HAT and P300, and boosted Ac-CoA content in the nucleus, promoting the acetylation levels of histone H3 and H4, ultimately producing antidepressant-like effects through increased histone acetylation and improved synaptic plasticity in the hippocampus.

Acetyl Coenzyme A Synthase 2 Acts as a Prognostic Biomarker Associated with Immune Infiltration in Cervical Squamous Cell Carcinoma

Chia-Jung Li et al.

Summary: Cellular metabolism plays a crucial role in cancer cell and immune cell viability and function. Tumors have a specialized metabolism different from normal cells to meet their anabolic demands, with ACSS2 as a key enzyme synthesizing acetyl-CoA. High expression of ACSS2 in CESC patients is associated with increased immune infiltration and poorer prognosis, indicating its potential as a diagnostic and prognostic biomarker.
