Acquired insights from the long-term surveillance of SARS-CoV-2 RNA for COVID-19 monitoring: The case of Monterrey Metropolitan Area (Mexico)

Diego A. Padilla-Reyes et al.

Summary: Wastewater-based epidemiology is an effective and cost-effective method for monitoring the spread of SARS-CoV-2, providing early warning of outbreaks in the population. The study found a correlation between viral load in wastewater and clinical cases, with wastewater surveillance able to provide a 2-7 day early warning. The detection rate of SARS-CoV-2 in wastewater was relatively low, with one detection for every 14,300 reported new cases. Prevalence estimates during the study period ranged from 0.02% to 4.6%. The application of wastewater-based epidemiology for COVID-19 surveillance is generally consistent and feasible, with room for improvement.

A deletion in the N gene of SARS-CoV-2 may reduce test sensitivity for detection of SARS-CoV-2

Huanyu Wang et al.

Summary: A 3 nucleotide deletion was found in a SARS-CoV-2-positive sample, causing impaired detection of the N1 target by RT-PCR using US CDC primer/probe sets. This finding highlights the importance of continued surveillance of SARS-CoV-2 mutations and assessment of their impact on diagnostic test performance.

Detection of SARS-CoV-2 RNA in wastewater, river water, and hospital wastewater of Nepal

Sarmila Tandukar et al.

Summary: This study reports the presence and reduction of SARS-CoV-2 in wastewater from wastewater treatment plants and sewer lines in Nepal. It found a high prevalence of SARS-CoV-2 in wastewater, indicating that biological treatments at the treatment plants are not enough to eliminate the virus. The study suggests that wastewater-based epidemiology is a useful tool for estimating COVID-19 cases in developing countries.

Persistence of Fecal Indicators and Microbial Source Tracking Markers in Water Flushed from Riverbank Soils

Jose S. Calderon et al.

Summary: This study evaluated the persistence and decay of different microbial indicators in contaminated riverbank soils. The findings showed that microbial indicators could still be detected in flush water months after contamination, with PMMoV persisting the longest and E. coli degrading the most rapidly. The study also highlighted the importance of using multiple human-associated fecal indicators to distinguish pollution from different microbial groups in water bodies.

Warish Ahmed et al.

Summary: The study found that 24-hour composite samples offer increased analytical sensitivity and decreased variability compared to 1-hour composite samples when monitoring wastewater, especially for pathogenic viruses with low infection rates within a community. This suggests that 24-hour composite samples are less likely to produce false negative results and erroneous public health information in wastewater-based epidemiology applications.

Decay of infectious SARS-CoV-2 and surrogates in aquatic environments

Laura Sala-Comorera et al.

Summary: The study found that the persistence of infectious SARS-CoV-2 in water was temperature dependent, with the virus remaining infectious for longer periods in colder temperatures. In contrast, viral RNA was relatively stable. The use of surrogate markers such as bacteriophages MS2 and PMMoV RNA showed significant differences in decay rates compared to SARS-CoV-2 RNA, limiting their effectiveness in assessing the persistence of the virus in the environment.
