Copper-catalyzed C-H/N-H annulation of enaminones and alkynyl esters for densely substituted pyrrole synthesis

Summary: In this study, a copper-catalyzed annulation of enaminones with alkynyl esters was developed for the synthesis of different pyrroles with a 2,3,4,5-tetrasubstituted structure. By using Cu(OAc)(2) as the catalyst, the synthesis of 2-vinyl and 2,3-dicarboxyl-functionalized pyrroles was achieved using terminal and internal alkynyl esters, respectively. The synthesis of 2-vinyl pyrroles represents the first example of accessing 2-vinyl substituted pyrroles via direct cascade reactions involving vinylation and pyrrole ring formation.

Catellani Reaction: An Enabling Technology for Vicinal Functionalization of Aryl Halides by Palladium(0)/Norbornene Cooperative Catalysis

The review focuses on the significant progress in such transformations and categorizes typical Catellani reactions into five parts based on the functionalization modes of ortho-C-H bonds: alkylation, arylation, amination, acylation, or thiolation.

Under the protection of a nitrogen atmosphere, tribenzotropones are synthesized by a [4 + 3] decarboxylative cyclization. Employing a palladium/O-2 system enables a [4 + 2] decarboxylative cyclization to assemble triphenylenes. Preliminary mechanistic studies suggest that the formation of triphenylenes involves a double decarboxylation.

The functionalization of C-H and N-H bonds led to the formation of a variety of tricyclic and tetracyclic sulfoximine derivatives with moderate to excellent yields under simple reaction conditions.
