Advances in the site-selective C-5, C-3 and C-2 functionalization of chromones via sp2 C-H activation

Anjitha Theres Benny et al.

Summary: This work focuses on the site-selective C-H activation of chromones at the C-5, C-3, and C-2 positions to introduce structural diversity to the chromone scaffold. The keto group of the chromone moiety acts as a directing group for selective functionalization at the C-5 position. Electrophilic coupling partners allow for C-H functionalization at the electron-rich C-3 position, while nucleophilic coupling partners enable C-H functionalization at the C-2 position. Direct functionalization methods offer a more atom-economical pathway for generating C-5, C-3, and C-2-substituted chromones compared to classical pre-functionalized reaction protocols.

Determining Essential Requirements for Fluorophore Selection in Various Fluorescence Applications Taking Advantage of Diverse Structure-Fluorescence Information of Chromone Derivatives

Yikun Chen et al.

Summary: This work explores the essential requirements for fluorophore selection in various fluorescence applications by assembling a library of chromone-derived fluorophores with diverse structure-fluorescence properties. The selection standard of a suitable fluorophore varies depending on the application: strong and constant fluorescence is preferred in fluorescence imaging, relatively lower fluorescence quantum yield favors detection sensitivity in the detection of bioactive species, and the key parameter in enzymatic assays employing fluorescence is the binding affinity between the fluorophore and the enzyme.
