African swine fever virus cysteine protease pS273R inhibits pyroptosis by noncanonically cleaving gasdermin D

Gaihong Zhao et al.

Summary: Research has found that the pyroptosis execution protein gasdermin D (GSDMD) is a new binding partner of ASFV-encoded protein S273R (pS273R). Further experiments showed that pS273R can cleave swine GSDMD, producing a shorter N-terminal fragment that does not trigger pyroptosis or inhibit ASFV replication. This discovery reveals a previously unrecognized mechanism involved in the inhibition of pyroptosis induced by ASFV infection.

MGF360-9L Is a Major Virulence Factor Associated with the African Swine Fever Virus by Antagonizing the JAK/STAT Signaling Pathway

Keshan Zhang et al.

Summary: This study identified the function of MGF360-9L protein during ASFV infection. MGF360-9L inhibits IFN-beta signaling through the degradation of STAT1 and STAT2 and is an important virulence gene of ASFV. These findings may guide the development of live attenuated ASFV vaccines.

African Swine Fever Virus F317L Protein Inhibits NF-κB Activation To Evade Host Immune Response and Promote Viral Replication

Jinping Yang et al.

Summary: African swine fever (ASF) is a highly contagious viral disease affecting pigs with a high mortality rate. The uncharacterized protein F317L of ASFV plays an antagonistic role against host innate immune response, impacting the NF-kappa B pathway activation and expression of proinflammatory cytokines.

African swine fever virus protein MGF-505-7R promotes virulence and pathogenesis by inhibiting JAK1-and JAK2-mediated signaling

Dan Li et al.

Summary: This study elucidates the mechanism by which the ASFV-encoded protein MGF-505-7R inhibits the JAK-STAT1 signaling pathway, resulting in reduced replication capacity of ASFV in the host. Deletion of MGF-505-7R attenuates the virulence and pathogenesis of ASFV, suggesting its potential as a strategy for developing vaccines against ASFV.

African Swine Fever Virus E120R Protein Inhibits Interferon Beta Production by Interacting with IRF3 To Block Its Activation

Huisheng Liu et al.

Summary: African swine fever is a devastating disease caused by ASFV, and the E120R protein of ASFV was identified as a key virulence factor that suppresses the host antiviral response by inhibiting interferon production. Targeting E120R could guide the development of ASFV live-attenuated vaccines.

Summary: The study identified interactions between ASFV MGF360-9L protein and numerous other proteins in porcine kidney cells, with implications in protein binding, metabolism, and immune response. Overexpression of certain proteins like PSMA3 and PSMC1 significantly promoted ASFV replication, shedding light on potential mechanisms of viral pathogenesis.

pMGF505-7R determines pathogenicity of African swine fever virus infection by inhibiting IL-1β and type I IFN production

Jiangnan Li et al.

Summary: The study revealed that ASFV infection inhibits the production of IL-1 beta and type I IFNs in porcine alveolar macrophages, highlighting the critical role of pMGF505-7R in this process. ASFV lacking the MGF505-7R gene showed reduced virulence in pigs and induced higher levels of IL-1 beta and IFN-beta production. The findings provide insights into the functions of ASFV-encoded pMGF505-7R and its impact on viral pathogenesis, potentially aiding in the development of antiviral strategies or vaccines to control ASF.
