Electrochemical One-Step Immunoassay Based on Switching Peptides and Pyrolyzed Carbon Electrodes

Jun-Hee Park et al.

Summary: Switching peptides were designed as reversible binding antibodies for different antigens and source animals. An electrochemical one-step immunoassay using switching peptides labeled with ferrocene was conducted for quantitative measurement of analytes. The feasibility of this method was assessed by analyzing electrochemical properties and the factors affecting the amperometry of switching peptides labeled with ferrocene. The applicability of the electrochemical immunoassay for medical diagnosis of human hepatitis B surface antigen was evaluated.

One-step immunoassay for food allergens based on screened mimotopes from autodisplayed FV-antibody library

Jeong Soo Sung et al.

Summary: This article presents a one-step immunoassay technique that utilizes mimotopes to detect target analytes. By screening mimotopes from an autodisplayed F-V-antibody library, the target analyte can be quantitatively detected without any washing steps. The CDR3 regions of the screened F-V-antibodies showed binding affinity to food allergens, which were synthesized into mimotopes for the corresponding food allergens. The one-step immunoassays for food allergens were successfully demonstrated without the need for immobilization of antibodies to a solid support.

Capacitive biosensor based on vertically paired electrodes for the detection of SARS-CoV-2

Jun-Hee Park et al.

Summary: Vertically paired electrodes (VPEs) with multiple electrode pairs were developed to enhance capacitive measurements. The sensitivity was improved by optimizing the electrode gap and number of electrode pairs. A conductive polymer layer was used for electrode fabrication, and the sensitivity enhancement was verified through modeling and experiments. The VPEs were successfully applied for SARS-CoV-2 nucleoprotein detection.

Switching-peptides for one-step immunoassay and its application to the diagnosis of human hepatitis B

Ji-Hong Bong et al.

Summary: In this study, switching-peptides were used to develop a one-step immunoassay for detecting antigen-antibody interactions without the need for additional antibody treatment or washing steps. The design principles of switching-peptides and the study of their interaction with antibodies using thermodynamic parameters demonstrated successful binding properties. The feasibility of using switching-peptides for quantitative one-step immunoassay was confirmed through HBsAg detection and statistical comparison with conventional ELISA results, showing promising results for practical applications.
