The CRISPR-Cas toolbox and gene editing technologies

Summary: Two type II-C Cas9 orthologs, Nm1Cas9 and Nm2Cas9, were used for efficient genome editing in plants with distinct PAM preferences, providing a reliable strategy to extend the scope of genome editing. Base editors developed from NmCas9s showed improved performance on the same targets, suggesting that engineering of NmCas9s or other type II-C Cas9s could offer useful alternatives for crop genome editing.

The optimized approach achieved higher editing frequencies compared to the original editor, Prime Editor 3, showing the potential to make previously non-editable target sites editable and expand the capabilities of prime editing in the future.
