Transcriptional and DNA Methylation Signatures of Subcutaneous Adipose Tissue and Adipose-Derived Stem Cells in PCOS Women

Adeline Divoux et al.

Summary: The study found that the gluteofemoral adipose tissue of PCOS patients showed lower expression of genes related to lipid accumulation and angiogenesis compared to abdominal adipose tissue, with an increase in preadipocyte number in both depots. RNA-seq pathway analysis demonstrated that ASCs derived from PCOS are pro-inflammatory in abdominal depot and exhibit a hypoxic signature in gluteofemoral depot, with lower expression of adipogenic genes. Additionally, a higher CpG methylation level was found in PCOS compared to control exclusively in gluteofemoral ASCs.

Non-coding RNAs in polycystic ovary syndrome: a systematic review and meta-analysis

Liangshan Mu et al.

Summary: A variety of genetic, environmental and epigenetic factors may play important roles in the pathogenesis of polycystic ovary syndrome (PCOS), but the exact cause remains unclear. Studies have shown that non-coding RNAs (ncRNAs) are involved in the occurrence and development of PCOS. Aberrant expression of ncRNAs in PCOS may lead to abnormal steroidogenesis, adipocyte dysfunction, altered ovarian cell proliferation and/or apoptosis, and have the potential to be used as diagnostic biomarkers.

lnc-MAP3K13-7:1 Inhibits Ovarian GC Proliferation in PCOS via DNMT1 Downregulation-Mediated CDKN1A Promoter Hypomethylation

Xueying Geng et al.

Summary: This study revealed a novel mechanism of DNA hypomethylation induced by lncRNAs, particularly lnc-MAP3K13-7:1, in patients with PCOS, leading to impaired proliferation of GCs. The overexpression of lnc-MAP3K13-7:1 inhibited DNMT1 expression, resulting in increased CDKN1A/p21 expression and cell cycle arrest in the G(0)/G(1) phase. This mechanism acts as a critical pathway in regulating GC growth and may provide potential therapeutic targets for PCOS-related infertility.

Multiomics Analysis Reveals Molecular Abnormalities in Granulosa Cells of Women With Polycystic Ovary Syndrome

Rusong Zhao et al.

Summary: In this study, genome-wide DNA methylation and lncRNA-miRNA-mRNA profiles were simultaneously detected in granulosa cells of PCOS cases and controls. Differential expressions of lncRNAs, miRNAs, mRNAs, and DMR-associated genes were found, with pathway analysis indicating their association with steroid biosynthesis and metabolic pathways. The study also identified known and new competing endogenous RNA (ceRNA) axes, most of which were related to steroid biosynthesis and metabolism. These findings emphasize the importance of considering reproductive and metabolic genes in the pathogenesis of PCOS.

Reduced stress-associated FKBP5 DNA methylation together with gut microbiota dysbiosis is linked with the progression of obese PCOS patients

Fu Chen et al.

Summary: PCOS patients exhibit higher levels of androgens and HOMA-IR, as well as lower FKBP5 DNA methylation, with differences in bacterial diversity and community compared to healthy controls. Significant changes are observed in bacterial genera and plasma metabolites in PCOS patients, indicating a potential link between stress responses and the progression of PCOS.
