Delivery of affordable and scalable encapsulated allogenic/autologous mesenchymal stem cells in coagulated platelet poor plasma for dental pulp regeneration

Ioannis Angelopoulos et al.

Summary: This study evaluated the potential of Platelet Poor Plasma (PPP) as a biomaterial for regenerative endodontics procedures (REPs). The study found that PPP scaffold was compatible with dental pulp stem cells (DPSC) and umbilical cord mesenchymal stem cells (UC-MSC), with differences in proliferation and angiogenic properties. Histological analysis showed the formation of dentin-like structure with MSC-PPP implants. This study provides evidence of a cost-effective scaffold for dental pulp regeneration.

Biomimetic mineralized microenvironment stiffness regulated BMSCs osteogenic differentiation through cytoskeleton mediated mechanical signaling transduction

Lu Chen et al.

Summary: Construction of biomimetic microenvironment is essential to understand the relationship between matrix mechanical cues and cell fate. Higher mechanical strength in the biomimetic mineralized microenvironment promoted osteogenic differentiation of stem cells through enhanced cytoskeletal assembly and expression of YAP and RUNX2. This study provides a promising material platform for bone tissue engineering and exploring the mechanism of biomimetic bone matrix mechanics on osteogenesis.

Leucine rich amelogenin peptide prevents ovariectomy-induced bone loss in mice

Naoto Haruyama et al.

Summary: Amelogenins, particularly LRAP, play important roles in bone turnover by regulating bone formation and osteoclastogenesis. Transgenic mice overexpressing LRAP showed enhanced bone formation, inhibited osteoclastogenesis, and resistance to bone loss, highlighting the potential therapeutic implications of LRAP in treating osteoporosis.

Chlorite oxidized oxyamylose differentially influences the microstructure of fibrin and self assembling peptide hydrogels as well as dental pulp stem cell behavior

Mostafa EzEldeen et al.

Summary: Tailored hydrogels mimicking the native extracellular environment have been studied for their potential in regenerative endodontics. This study evaluated the effects of a chemokine-binding and antimicrobial polymer, COAM, on the microstructural properties of fibrin and SAP hydrogels, as well as the behavior of hDPSCs. The inclusion of COAM did not alter the microstructure of fibrin hydrogels significantly, but affected the SAP hydrogels leading to fiber aggregation. Cell viability and attachment were higher in fibrin hydrogels compared to SAP hydrogels, suggesting the former as a promising carrier for COAM in endodontic regeneration.

Esteban Astudillo-Ortiz et al.

Summary: The study investigated an injectable hyaluronic acid-based hydrogel system enriched with platelet lysate for endodontic regeneration. The hydrogels showed suitable working and setting times, a porous microstructure, and supportive properties for cell migration towards dentin surfaces, indicating great clinical potential for endodontic regenerative therapies.

Evaluating the potential of an amelogenin-derived peptide in tertiary dentin formation

Xiu Peng et al.

Summary: Several novel biomaterials have been developed to induce tertiary dentin formation for dental pulp capping. QP5, an amelogenin-based peptide, was found to enhance migration and mineralization of human dental pulp cells, with little effect on cell viability. In vivo results in rat models showed that QP5 as a pulp capping agent promoted the formation of tertiary dentin more prominently compared to the negative control group.

Zohre Mousavi Nejad et al.

Summary: The PCL/BG and PCL/HyA scaffolds fabricated by 3D printing technology showed great potential for promoting dentin and pulp tissue regeneration, with the former exhibiting higher bioactivity and surface roughness, and the latter enhancing cell adhesion and hydrophilicity. Gene expression results demonstrated increased expression of odontogenic markers with both scaffolds, indicating their suitability for hDPSC adhesion and odontogenic differentiation.
