An RNAi screen of the kinome in epithelial follicle cells of the Drosophila melanogaster ovary reveals genes required for proper germline death and clearance

Diane P. Lebo et al.

Summary: This study identified novel genes required for clearance by nonprofessional phagocytes in the Drosophila ovary, revealing pathways not previously implicated in phagocytosis. Interestingly, several genes were found to affect both the clearance process in phagocytes and non-autonomously influence germline cell death. This unbiased kinase screen provided new avenues for further exploration and investigation.

Differential activation of JAK-STAT signaling reveals functional compartmentalization in Drosophila blood progenitors

Diana Rodrigues et al.

Summary: This study reveals the heterogeneity of posterior progenitors (PP) in the Drosophila larval lymph gland and identifies molecular-genetic tools for studying this cell population. Functional analysis shows that PP resist differentiation upon immune challenge, promoting survival in a JAK-STAT-dependent manner, contrasting with the response of anterior lobe progenitors (AP).

Polarization of Macrophages in Insects: Opening Gates for Immuno-Metabolic Research

Adam Bajgar et al.

Summary: Research suggests that in the acute phase of bacterial infection, signaling factors released by bactericidal macrophages induce insulin resistance, which may act as a metabolic adaptive mechanism to support immune response. While this metabolic adaptation allows macrophages to combat pathogens efficiently, it also increases their nutritional demands, highlighting the importance of coordinating cellular metabolism and systemic metabolic changes during infections.

Identification of functionally distinct macrophage subpopulations in Drosophila

Jonathon Alexis Coates et al.

Summary: Vertebrate macrophages are highly heterogeneous, and recent studies have revealed the presence of subpopulations within Drosophila plasmatocytes, showing functional differences and varying in abundance across development. Enhancer analysis identified candidate genes regulating plasmatocyte behavior and exposure to increased apoptotic cell death levels modulated subpopulation cell numbers. These findings demonstrate macrophage heterogeneity in Drosophila and provide insights into subpopulation specification and function.

Diet-regulated production of PDGFcc by macrophages controls energy storage

Nehemiah Cox et al.

Summary: This study identified a macrophage-dependent mechanism involving the PDGFcc ortholog that controls lipid storage in adipose tissues of mice. Production of PDGFcc is regulated by diet and blocking PDGFcc leads to redirection of excess lipids toward thermogenesis in brown fat.

Drosophila as a Model to Study Cellular Communication Between the Hematopoietic Niche and Blood Progenitors Under Homeostatic Conditions and in Response to an Immune Stress

Ismael Morin-Poulard et al.

Summary: In adult mammals, blood cells are formed from hematopoietic stem progenitor cells, which are controlled by a complex cellular microenvironment called niche. Drosophila melanogaster is a powerful model organism to decipher the mechanisms controlling hematopoiesis, due to its limited number of blood cell lineages and the conservation of genes and signaling pathways throughout bilaterian evolution. Hematopoiesis in Drosophila occurs in two waves – during embryogenesis and at larval stages, regulated by distinct niches in different hematopoietic sites.

A Conserved Role for Asrij/OCIAD1 in Progenitor Differentiation and Lineage Specification Through Functional Interaction With the Regulators of Mitochondrial Dynamics

Arindam Ray et al.

Summary: Mitochondria are dynamic organelles that play a crucial role in blood stem and progenitor cell biology. The protein Asrij/OCIAD1 regulates mitochondrial morphology, energy metabolism, and cell differentiation in both human embryonic stem cells and Drosophila hematopoiesis. Studies in both model systems suggest a conserved role for Asrij in linking mitochondrial dynamics and progenitor differentiation.
