Osteoarthritis year in review 2021: epidemiology & therapy

J. G. Quicke et al.

Summary: This article reviews research themes and individual studies in the clinical osteoarthritis field, including COVID-19, disease burden, occupational risk, prediction models, etc. Important findings include the impact of non-steroidal anti-inflammatory drugs on COVID-19 related deaths, the increasing incidence rate of OA, and the relationship between cartilage thickness loss and pain.

Platelet-Rich Plasma Versus Hyaluronic Acid for Knee Osteoarthritis: A Systematic Review and Meta-analysis of Randomized Controlled Trials

John W. Belk et al.

Summary: This meta-analysis suggests that platelet-rich plasma (PRP) injections may be more effective than hyaluronic acid (HA) injections for treating knee osteoarthritis, and that leukocyte-poor PRP could be superior to leukocyte-rich PRP in terms of clinical outcomes.

Regenerative Potential of Mesenchymal Stem Cells for the Treatment of Knee Osteoarthritis and Chondral Defects: A Systematic Review and Meta-analysis

Bhargavi Maheshwer et al.

Summary: This study conducted a systematic review and meta-analysis on the effects of mesenchymal stem cells (MSCs) on cartilage regeneration, pain, and function. The results showed that MSC treatment significantly improved functional outcomes but did not have a significant impact on pain improvement. Furthermore, MSC treatment led to a significant increase in cartilage volume, but not in cartilage quality.

Cell-based therapy in soft tissue sports injuries of the knee: a systematic review

Nardeen Kader et al.

Summary: The evidence for the effectiveness of cell-based therapies in soft-tissue sports injuries of the knee is currently lacking high-quality support. There is a need for more standardized protocols and reporting, as well as further basic scientific research and large clinical trials to better understand the potential of MSCs and improve their application in clinical settings.

Assessment of safety and efficacy of intra-articular injection of stromal vascular fraction for the treatment of knee osteoarthritis-a systematic review

Saseendar Shanmugasundaram et al.

Summary: Intra-articular injection of SVF is shown to be a safe and effective treatment for knee osteoarthritis, with improvements in pain, range of motion, and minimal donor-site morbidity. Further comparative Level I studies are needed to evaluate the use of adjuvants with SVF and compare it with other regenerative therapies.

Enhanced anti-inflammatory effects of mesenchymal stromal cells mediated by the transient ectopic expression of CXCR4 and IL10

Rosario Hervas-Salcedo et al.

Summary: The transient co-expression of CXCR4 and IL10 enhances the therapeutic potential of MSCs in a local inflammation mouse model, indicating that mRNA-modified cells may be a new step towards more efficient cell therapies for inflammatory diseases.

Summary: MSCs are widely used in advanced therapies for various purposes, mainly related to inflammatory processes. Despite their demonstrated efficacy in preclinical models and early clinical trials, there are still controversies regarding their specific phenotype, delivery methods, and varied cell products. Strategies for bioengineering MSCs are being explored to enhance their properties and functional capabilities.

Meta-Analysis of Adipose Tissue Derived Cell-Based Therapy for the Treatment of Knee Osteoarthritis

Nikhil Agarwal et al.

Summary: Cell-based therapies derived from adipose tissue, specifically AMSC therapy and SVF therapy, have been shown to be safe and effective treatments for knee OA in old adults, reducing pain and improving knee function over time.

Is Culture Expansion Necessary in Autologous Mesenchymal Stromal Cell Therapy to Obtain Superior Results in the Management of Knee Osteoarthritis?-Meta-Analysis of Randomized Controlled Trials

Sathish Muthu et al.

Summary: This study aimed to analyze the impact of cultured expansion of autologous mesenchymal stromal cells (MSCs) in the management of knee osteoarthritis. The results showed that culture expanded MSCs had better improvement at six months, while uncultured MSCs demonstrated significant improvement at 12 months in various outcomes. Adipose-derived uncultured MSCs outperformed culture-expanded MSCs in functional outcomes at both short and long term, suggesting that culture expansion of MSCs may not be necessary for superior results in knee osteoarthritis management.
