New contributions to the phylogeny of the ciliate class Heterotrichea (Protista, Ciliophora): analyses at family-genus level and new evolutionary hypotheses

Yong Chi et al.

Summary: This study utilized both morphological and molecular data to infer phylogenetic relationships for the class Heterotrichea, proposing new evolutionary hypotheses. Key findings include discrepancies between molecular and morphological evidence in certain relationships, as well as uncertainty in the systematic positions of several families. Monophyly of each genus in the study was supported by molecular phylogenetic trees, with a few exceptions.

A Polycomb repressive complex is required for RNAi-mediated heterochromatin formation and dynamic distribution of nuclear bodies

Jing Xu et al.

Summary: In this study, the EZL1 complex in the protist Tetrahymena thermophila was characterized, revealing its essential role in histone methylation, heterochromatin formation, and genome rearrangement. The interaction between the EZL1 complex and EMA1, as well as PDD1, reinforces its chromatin association and contributes to the dynamic distribution of Polycomb bodies in Tetrahymena development. These findings provide a molecular mechanism linking RNAi and Polycomb repression, ultimately regulating nuclear bodies and genome organization.

Taxonomy and molecular phylogeny of four karyorelictid species belonging to the genera Apotrachelocerca and Tracheloraphis (Protozoa: Ciliophora), with descriptions of two new species

Mingzhen Ma et al.

Summary: Taxonomy and phylogeny of four trachelocercid ciliates, two of which are new species, isolated from marine habitats in Qingdao, China were studied. The new species were distinguished from their closely related congeners by differences in cortical granules and SSU rDNA sequence. Removal of one species from a complex was suggested based on the presence of certain cortical granules. Phylogenetic analyses revealed specific groupings among the species studied.

The Compact Macronuclear Genome of the Ciliate Halteria grandinella: A Transcriptome-Like Genome with 23,000 Nanochromosomes

Weibo Zheng et al.

Summary: This study reveals that the genome of Halteria grandinella is composed of gene-sized nanochromosomes with minimal non-genic regions, challenging traditional understanding of chromosomal structure. The transcription dynamics of Halteria are influenced by factors such as non-uniform nanochromosome copy numbers and extremely short untranslated regions. These findings broaden our understanding of ciliate biology and the evolution of unicellular eukaryotes.

New contribution to epigenetic studies: Isolation of micronuclei with high purity and DNA integrity in the model ciliated protist, Tetrahymena thermophila

Lili Duan et al.

Summary: This study presents a reliable and quick method for isolating high-purity and DNA-integrity micronuclei in Tetrahymena. Various factors were examined to optimize micronuclei purification, and a more accurate method for detecting nuclear contamination was established. This research will facilitate further epigenetic studies in Tetrahymena.

Chromosome organization and gene expansion in the highly fragmented genome of the ciliate Strombidium stylifer

Chao Li et al.

Summary: Chromosomes are well-organized carriers of genetic information in eukaryotes, with Spirotrichea ciliates featuring nanochromosomes predominantly carrying only one gene. The copy number of nanochromosomes in Strombidium stylifer lacks precise regulation.

Large-scale phylogenomic analysis provides new insights into the phylogeny of the class Oligohymenophorea (Protista, Ciliophora) with establishment of a new subclass Urocentria nov. subcl

Chundi Wang et al.

Summary: This study sequenced the genomes and/or transcriptomes of six non-culturable oligohymenophoreans using single-cell sequencing techniques. Phylogenomic analysis with expanded taxon sampling of 85 taxa revealed the evolutionary relationships within Oligohymenophorea and proposed new classifications for Urocentrids and Paranassula. The study also provided insights into the early divergence of Peritrichia and the evolutionary usage of stop codons among oligohymenophoreans.
