Microbial Metabolite Inspired β-Peptide Polymers Displaying Potent and Selective Antifungal Activity

Summary: With the quick increase of serious invasive fungal infections and the limited antifungal drugs available, there is an urgent need for potent and selective antifungal agents. Inspired by microbial metabolites, the authors have designed and obtained poly(DL-diaminopropionic acid) (PDAP), a potent antifungal agent that kills fungal cells by penetrating the cytoplasm, generating reactive oxygen, and inducing fungal apoptosis. PDAP displays strong antifungal activity against Candida albicans, negligible hemolysis and cytotoxicity, and no susceptibility to antifungal resistance. In addition, PDAP effectively inhibits fungal biofilm formation and eradicates mature biofilms. In vivo studies have shown that PDAP is safe and effective in treating fungal keratitis, highlighting its potential as a promising new antifungal agent.

Critical effect of biodegradation on long-term microplastic weathering in sediment environments: A systematic review

Summary: This article reviews the enzyme induced biodegradation processes of microplastics in sediment environments, exploring the factors influencing plastic degradation such as polymer type, molecular weight, crystallinity, and hydrophobicity of microplastics, as well as sediment conditions like sediment type, temperature, pH, salinity, and oxygen content. The study also discusses how microbial abundance, enzyme concentrations, and enzyme activities can alter the biodegradation ratios. The findings presented in this review contribute to a better understanding of the formation process, fate, and potential ecological risks associated with aged microplastics in sediment environments.

Cyclic Poly(α-peptoid)s by Lithium bis(trimethylsilyl)amide (LiHMDS)-Mediated Ring-Expansion Polymerization: Simple Access to Bioactive Backbones

Summary: This study presents a simple method for the synthesis of cyclic poly(alpha-peptoids) using LiHMDS promoted ring-expansion polymerization (REP) from N-alkylated-N-carboxyanhydrides (NNCA). The new method allows the use of lysine-like monomers in REP to design bioactive macrocycles with potential pharmaceutical applications against Clostridioides difficile. This novel approach offers a more efficient and straightforward route compared to traditional methods.

Summary: This study demonstrates a living and controlled copolymerization to prepare sequence-tunable poly-alpha/beta-peptides, showing a prominent solvent-driven characteristic. It provides random-like copolymers of poly-alpha/beta-peptides in THF and block-like copolymers of poly-alpha/beta-peptides in a mixed solvent of CHCl3/H2O (95/5, v/v), opening new avenues for sequence-tunable polymerization and enabling facile synthesis of proteolysis tunable poly-alpha/beta-peptides for diverse applications.

Summary: Sulfated poly-amido-saccharides (sulPASs) have been reported as effective heparin mimetics, inhibiting intrinsic pathway of coagulation with activity positively correlated to molecular weight and degree of sulfation. They are not degraded by heparanases and non-hemolytic, showing potential as synthetic anticoagulants with comparable bleeding risk to heparin in in vivo murine model.
