In Vivo Pharmacodynamic Method to Assess Complement C5a Receptor Antagonist Efficacy

Summary: C5a receptor 1 inhibitors have been studied as potential therapeutic targets for autoimmune and inflammatory diseases, with PMX53 and JPE-1375 identified as drug candidates. A robust in vivo pharmacodynamic model was developed in mice, showing that both PMX53 and JPE-1375 have similar in vivo efficacy in inhibiting C5aR1-mediated responses, but PMX53 has a longer duration of action. Pharmacokinetic analysis revealed rapid distribution and elimination of both compounds, with PMX53 having a longer half-life, allowing for the development of an accurate pharmacokinetic/pharmacodynamic model.

A validated quantitative method for the assessment of neuroprotective barrier impairment in neurodegenerative disease models

Summary: The blood-brain barrier (BBB) and blood-spinal cord barrier (BSCB) are specialized structures that limit molecule entry and maintain homeostasis within the central nervous system. This study developed and validated a quantitative method for assessing BBB and BSCB integrity using sodium fluorescein. The method revealed changes in permeability in models of neurodegenerative diseases and could contribute to understanding the role of these neuroprotective barriers in neurodegeneration processes.

All patients received cyclophosphamide or rituximab. The safety and clinical effects of avacopan beyond 52 weeks were not addressed in the trial.
