One-Pot Preparation of Highly Dispersed Second Near-Infrared J-Aggregate Nanoparticles Based on FD-1080 Cyanine Dye for Bioimaging and Biosensing

Caixia Sun et al.

Summary: Bioimaging and biosensing in the second near-infrared (NIR-II) window have great potential due to their high resolution and penetration depth. However, the synthesis of hydrophilic organic contrast agents with absorbance and emission wavelengths beyond 1300 nm remains challenging. This study demonstrates the use of J-aggregation to achieve red-shifted organic dyes in the NIR-II window and improve their hydrophilicity. FD-1080 J-aggregates (FD-J) with absorbance of 1360 nm and emission of 1370 nm were successfully synthesized, enabling real-time imaging of mice brain and hindlimb vasculatures beyond 1500 nm. In addition, these aggregates were used as fluorescence sensors to detect inflammation in living mice.

Targeted multicolor in vivo imaging over 1,000 nm enabled by nonamethine cyanines

Venu G. Bandi et al.

Summary: Through rational design, red-shifted, bioconjugatable heptamethine cyanine dyes were developed for multiplexed in vivo imaging in the shortwave-infrared/near-infrared-II region. The use of wavelengths between 1,000 and 2,000 nm enables high-resolution in vivo imaging at depths not possible with conventional optical wavelengths. Recent progress has shown a lack of bioconjugatable probes for imaging in the shortwave-infrared or near-infrared (NIR)-II range.
