Somatic Mutations Enriched in Cis-Regulatory Elements Affect Genes Involved in Embryonic Development and Immune System Response in Neuroblastoma

Vito Alessandro Lasorsa et al.

Summary: This study focuses on noncoding cis-regulatory variants in neuroblastoma and identifies somatic mutations in regulatory elements that play a role in tumorigenesis. The findings highlight the importance of assessing these variants and long-range interactions in understanding the drivers of neuroblastoma.

Single-cell transcriptional changes associated with drug tolerance and response to combination therapies in cancer

Alexandre F. Aissa et al.

Summary: Tyrosine kinase inhibitors have clinical effectiveness in treating certain subsets of cancers with receptor tyrosine kinase somatic mutations, but limited duration of response and development of drug resistance are common issues. Single-cell RNA sequencing revealed multiple cancer cell subpopulations with epigenetic changes and variable therapeutic sensitivity. Overrepresented gene ontologies associated with drug tolerance include epithelial-to-mesenchymal transition, drug metabolism, and vesicle transport. Combination therapy targeting specific drug tolerant cell subpopulations shows promise in inhibiting drug resistance.

Metallothionein-3 promotes cisplatin chemoresistance remodelling in neuroblastoma

Miguel Angel Merlos Rodrigo et al.

Summary: This study revealed that up-regulation of metallothionein-3 (hMT3) in neuroblastoma cells was associated with increased cisplatin resistance, indicating a potential cause for tumor relapse. Multiple biological pathways related to apoptosis, transport, proteasome, and cellular stress were implicated in cisplatin resistance in hMT3-overexpressing neuroblastoma cells.

Tumor Heterogeneity: A Great Barrier in the Age of Cancer Immunotherapy

Nader El-Sayes et al.

Summary: Tumor heterogeneity remains a major obstacle in successful cancer treatment, arising from changes in the tumor microenvironment and leading to the presence of different malignant subpopulations within a single tumor mass. Therapeutic interventions can increase selective pressure towards resistant subpopulations, often causing relapse in previously responsive patients. Strategies such as multi-therapy regimens are being employed to combat intrinsic resistance mechanisms, especially in the field of cancer immunotherapy, where antigen escape and immunosuppression are common forms of acquired resistance.

Single-cell transcriptomics of human embryos identifies multiple sympathoblast lineages with potential implications for neuroblastoma origin

Polina Kameneva et al.

Summary: Single-cell transcriptome profiling of human embryonic sympathoadrenal tissues reveals developmental transitions and suggests that intra-adrenal sympathoblasts from Schwann cell precursors may be potential neuroblastoma cell origins. The majority of extra-adrenal sympathoblasts appear to arise from migratory neural crest cells, while intra-adrenal sympathoblasts seem to be directly derived from nerve-associated Schwann cell precursors. This process persists during several weeks of development within the large intra-adrenal ganglia-like structures, potentially serving as reservoirs of originating cells in neuroblastoma.

Plasticity in Neuroblastoma Cell Identity Defines a Noradrenergic-to-Mesenchymal Transition (NMT)

Margot Gautier et al.

Summary: Neuroblastoma, a common extracranial pediatric solid tumor, exhibits significant clinical heterogeneity, with high-risk tumors having poor overall survival. Recent studies have identified two distinct cell identities in neuroblastoma - noradrenergic (NOR) and mesenchymal (MES) - with important differences in chemoresistance and invasive properties. Understanding the transdifferentiation between these two cell states is crucial for improving therapeutic strategies for neuroblastoma patients.
