Adverse postnatal developmental effects in offspring from humidifier disinfectant biocide inhaled pregnant rats

Jinsoo Lee et al.

Summary: Polyhexamethylene guanidine phosphate (PHMG-P), a primary biocide in humidifier disinfectants, can cause fatal pulmonary disease in Korea. Inhalation of PHMG-P during pregnancy adversely affects maternal health and embryo-fetal development. Prenatal exposure to PHMG-P results in increased perinatal death rates, decreased viability index, lower birth weight, and systemic toxicities in offspring. These findings suggest that prenatal PHMG-P exposure has long-term effects on offspring health and could impact human risk assessment.

Summary: This study investigated the adverse effects of CMIT/MIT exposure on zebrafish embryos at various developmental stages, revealing mechanisms such as spinal curvature, pericardial edema, altered heart rates, global DNA hypermethylation, and significant changes in locomotion behavior. Proteomics analysis indicated that suppression of cardiac muscle contractions and energy metabolism were potential underlying mechanisms of the adverse effects caused by CMIT/MIT. The study identified < 6 hpf as the most susceptible exposure window, followed by < 48 hpf for CMIT/MIT.

Summary: The study found that repeated exposure to low doses of PHMG-P induces asthma-like symptoms, possibly through modulation of the Th17 signaling pathway rather than the Th2 pathway. The asthmatic airway inflammation induced by PHMG-P showed a distinct pattern from typical allergic asthma, characterized by Th2-low, Th17-related, IgE-independent, and mixed granulocytic features.

Hyeonsu Ryu et al.

Summary: The lung disease caused by exposure to chemical substances in humidifier disinfectants has been identified in Korea. Several researchers found that exposure classification using questionnaires may not correlate with clinical severity classes determined through diagnosis, suggesting the lack of correlation may be due to misclassification in exposure assessment. This study revealed exposure misclassification and proposed a questionnaire-based exposure assessment methodology to overcome limitations in past exposure assessments.

Summary: This study investigates the mechano-stress responses of human bronchial epithelial cells to two different biocides and demonstrates the versatility of real-time imaging-based biomechanical analysis. While inconsistent average responses were observed in the treated cells, a higher degree of dispersion in the data set was found in cells treated with CMIT/MIT. The study sheds light on the comparative understanding of less studied substance CMIT/MIT in relation to more studied substance PHMG, contributing to more strategic risk management planning in toxicological accidents involving these ingredients.
