Role of Hydrotropes in Sparingly Soluble Drug Solubilization: Insight from a Molecular Dynamics Simulation and Experimental Perspectives

Rabindranath Paul et al.

Summary: The therapeutic efficacy of drug molecules depends on their bioavailability and solubility, with over 70% of formulated drugs showing limited effectiveness due to low water solubility. One technique to enhance water solubility is through hydrotropy, which has high industrial demand due to its cost-effectiveness and environmentally friendly nature. This feature article provides a clear introduction, detailed insights, applications in the pharmaceutical industry, and future directions of hydrotropic solubilization, serving as a valuable resource for researchers in the field.

Ion Size Dependences of the Salting-Out Effect: Reversed Order of Sodium and Lithium Ions

Hiroyuki Katsuto et al.

Summary: The general trend of the salting-out effect on hydrophobic solutes in aqueous solution is that smaller ions reduce the solubility of hydrophobes more effectively, with Li+ being an exception as it has weaker effect compared to Na+. Molecular dynamics simulations revealed that the Setschenow coefficient is correlated with the packing fraction of salt solution and the partial molar volume of an ion, explaining the reversed order in the cation series.

Cooperativity in micellar solubilization

Seishi Shimizu et al.

Summary: Sudden solubilization occurs widely below or around the critical micelle concentration of surfactants, with solutes inducing micellization and increasing the aggregation number of surfactants. Enhanced cooperativity in micellization upon solubilization is driven by surfactant self-association enhanced by solutes, forming a universal mechanism for cooperative solubilization common to hydrotropes and surfactants at low concentrations.

The impact of the counterion in the performance of ionic hydrotropes

Dinis O. Abranches et al.

Summary: The efficiency of an ionic hydrotrope increases with the hydrophobicity of its counterion, challenging the common belief that ionic hydrotropes should have small, densely charged counterions like sodium or chloride.
