Asymmetric Cascade Aza-Henry/Lactamization Reaction in the Highly Enantioselective Organocatalytic Synthesis of 3-(Nitromethyl)isoindolin-1-ones from α-Amido Sulfones

Lorenzo Serusi et al.

Summary: The asymmetric synthesis of novel 3-substitued isoindolinones was achieved using the asymmetric nitro-Mannich reaction and the in situ cyclization reaction. High enantioselectivities and yields were obtained in the presence of a neutral bifunctional organocatalyst. The investigation of the reactivity of the obtained products also revealed the potential for further functionalization.

Base-Promoted Cascade Reactions for the Synthesis of 3,3-Dialkylated Isoindolin-1-ones and 3-Methyleneisoindolin-1-ones

Antonio Macchia et al.

Summary: Cascade reactions of ortho-carbonyl-substituted benzonitriles with ((chloromethyl)sulfonyl)benzenes as pronucleophiles can lead to new isoindolin-1-ones with a tetrasubstituted C-3 position or (Z)-3-(sulfonyl-methylene)isoindolin-1-ones. These reactions are carried out under mild conditions without the need for metal catalysis, promoted only by environmentally benign K2CO3, showing potential applications for the synthesis of aristolactam natural products.

Synthesis of 3,3-Dialkyl-Substituted Isoindolinones Enabled by Nickel-Catalyzed Reductive Dicarbofunctionalization of Enamides

Ke Fang et al.

Summary: A novel nickel-catalyzed reaction method for the synthesis of 3,3-dialkyl-substituted isoindolinone frameworks from unactivated alkyl iodides and 1,1-disubstituted enamides has been reported. The method exhibits good functional group tolerance and enantioselectivity through the use of commercially available chiral Bn-Biox ligand.
