A multicolor and ratiometric fluorescent sensing platform for metal ions based on arene-metal-ion contact

Summary: A fluorescent sensing platform based on arene-metal-ion contact was developed for multicolor and ratiometric detection of metal ions across a broad wavelength range. The flexible molecular design allows for the detection of various metal ions with emission red-shifts. X-ray crystallography and theoretical calculations revealed the mechanism behind the emission red-shift induced by the coordinated metal ions. The probes showed promising differential sensing capabilities and applicability for ratiometric imaging of metal ions under live-cell conditions.

Summary: Near-infrared (NIR) fluorescent molecules are important for visualising biological processes, with P=O-substituted phospha-xanthene (POX) dyes being a promising scaffold due to their NIR emission and photostability. The development of POXs bearing a 4-ethynylphenyl group on the phosphorus atom allows for functionalisation with azide-tagged biomolecules via late-stage Cu-catalysed azide/alkyne cycloaddition (CuAAC), enabling target-selective labelling. A sophisticated NIR probe has been designed to have a bell-shaped pH-response and assess endosomal maturation, demonstrating the practical utility of functionalised POXs.
