The Putative Role of mTOR Inhibitors in Non-tuberous Sclerosis Complex-Related Epilepsy

Hannah E. Goldstein et al.

Summary: Research suggests that mTOR inhibitors may be an effective treatment for refractory epilepsy, particularly for children who have not responded to surgical treatment. Rapamycin and everolimus have been shown to reduce seizure frequency and have reasonable safety and tolerability. Investigating the treatment role of mTOR inhibitors in non-TSC epilepsy is a logical next step based on current evidence.

Convergent and Divergent Mechanisms of Epileptogenesis in mTORopathies

Lena H. Nguyen et al.

Summary: Hyperactivation of mTORC1 due to gene mutations along the PI3K-mTOR pathway and the GATOR1 complex leads to neurodevelopmental disorders and epilepsy. Despite converging impacts on mTORC1 activity, different gene variants may contribute to epilepsy through both mTORC1-dependent and -independent mechanisms.
