Insights into the binding of morin to human γD-crystallin

Rodrigo Aguayo-Ortiz et al.

Summary: This study identifies the binding site of flavonoid morin (MOR) on human gamma D-crystallin (H gamma DC) and confirms its stabilizing effect on the structure of H gamma DC. This is of significant importance for the discovery of new H gamma DC aggregation inhibitors.

Effects of the Bark Resin Extract of Garcinia nigrolineata on Chronic Stress-Induced Memory Deficit in Mice Model and the In Vitro Monoamine Oxidases and β-Amyloid Aggregation Inhibitory Activities of Its Prenylated Xanthone Constituents

Charinya Khamphukdee et al.

Summary: The study found that the bark resin extract of Garcinia nigrolineata has a therapeutic effect on learning and memory deficits in UCMS mice, reversed by reducing oxidative stress in the brain. Additionally, the prenylated xanthones isolated from the bark resin showed inhibitory activities against Aβ aggregation and MAOs.

Elucidation of molecular interactions between human γD-crystallin and quercetin, an inhibitor against tryptophan oxidation

Shiwani Rana et al.

Summary: Different post-translational modifications of eye lens crystallin proteins play a role in the development of cataract, with oxidative modification of tryptophan residues by quercetin inhibiting the formation of NFK associated with cataractogenesis. The interactions between quercetin and HGD were studied using fluorescence techniques, indicating energy transfer and involvement of hydrogen bonding and van der Waals interactions. Experimental observations were supported by docking results, confirming the inhibition of oxidation by quercetin.

Cataract-causing mutations L45P and Y46D promote γC-crystallin aggregation by disturbing hydrogen bonds network in the second Greek key motif

Chenxi Fu et al.

Summary: Congenital cataracts caused by genetic disorders are the primary cause of child blindness across the globe. This study investigated two mutations of gamma C-crystallin in Chinese families with nuclear congenital cataracts, revealing that these mutations destabilized the protein and increased aggregation propensity under environmental stress.

Imbalances in the eye lens proteome are linked to cataract formation

Philipp W. N. Schmid et al.

Summary: Analysis of lenses from different mouse strains with early-onset cataracts due to mutations in alpha-, beta-, or gamma-crystallin proteins reveals that the mutant crystallins are unstable in vitro, reduced in levels within the lens, and do not accumulate in the water-insoluble fraction. Precipitation of other crystallin proteins, including alpha-crystallins, suggests that imbalance in the lenticular proteome and altered crystallin interactions are the basis for cataract formation, rather than the aggregation propensity of the mutant crystallins.

Chemical Properties Determine Solubility and Stability in βγ-Crystallins of the Eye Lens

Megan A. Rocha et al.

Summary: Beta gamma-crystallins are essential structural and refractive proteins in the vertebrate eye lens, crucial for maintaining lens transparency. Mutations, post-translational modifications, and metal ion binding can lead to protein aggregation, reduced solubility, and opacity. Experimental techniques such as dye-binding assays and fluorescence are used to assess crystallin conformation when high-resolution structures are unavailable.

Noncovalent molecular interactions between antineoplastic drug gemcitabine and a carrier protein identified through spectroscopic and in silico methods

Mohd Sajid Ali et al.

Summary: The noncovalent molecular interactions between hen egg white lysozyme (HEWL) and the antineoplastic drug gemcitabine were studied using spectroscopic techniques and in silico approaches. The interaction was dominated by hydrophobic forces with a possible contribution of hydrogen bonding. Gemcitabine induced slight changes in the secondary structure of HEWL and was inferred to bind in a large hydrophobic cavity of the protein.
